间隙性连接蛋白37对动脉粥样硬化中单核巨噬细胞电流密度的影响

【摘要】:目的国内外关于间隙性连接蛋白37(Cx37)基因改变巨噬细胞内外的电流密度,使得动脉粥样硬化进一步发展的研究甚少。文中旨在观察Cx37对动脉粥样硬化中巨噬细胞电流密度的影响。方法选取30只Wistar大鼠,Cx37+(保留Cx37基因)、Cx37-(沉默Cx37基因)各15只。采用高脂饮食建立动脉粥样硬化模型;根据标本采集部位分为4组:Cx37+斑块组、Cx37-斑块组(取自斑块)及Cx37+血液组、Cx37-血液组(取自血液),用RT-PCR检测Cx37+大鼠不同部位Cx37的表达;分离Cx37+、Cx37-大鼠血液中及斑块处单核巨噬细胞并培养,用全细胞记录法测定电流密度并进行比较。结果 Cx37+大鼠斑块中单核巨噬细胞中Cx37的相对表达水平显著高于血液[(1.1±0.02)vs(0.6±0.03)],差异有统计学意义(P=0.010)。Cx37+、Cx37-大鼠均行能谱CT检测鼠颈动脉斑块检测,验证了动物模型的成功建立。80、120、160 ms时,Cx37+斑块组电流密度[(0.61±0.06)、(0.67±0.07)、(0.91±0.03)A/cm~2]显著高于Cx37+血液组[(0.49±0.02)、(0.61±0.03)、(0.67±0.02)A/cm~2]、Cx37-斑块组[(0.48±0.02)、(0.60±0.02)、(0.64±0.02)A/cm~2]、Cx37-血液组[(0.49±0.02)、(0.59±0.02)、(0.64±0.02)A/cm~2];200、240、320 ms时亦显著高于Cx37+血液组、Cx37-斑块组、Cx37-血液组(P0.05)。结论 Cx37对动脉粥样硬化中单核巨噬细胞斑块处电流密度的影响比外周血液更为显著,可促使巨噬细胞继续活化,动脉粥样硬化进一步发展。
[Abstract]:Objective to investigate the effect of gap junction protein 37 (Cx37) gene on the current density of macrophages and make the further development of atherosclerosis rare. The aim of this study was to observe the effect of Cx37 on the current density of macrophages in atherosclerosis. Methods 30 Wistar rats with Cx37 (Cx37 gene) and Cx37- (silent Cx37 gene) were selected. Atherosclerosis model was established by high-fat diet. The samples were divided into four groups: Cx37 plaque group, Cx37- plaque group (taken from plaque) and Cx37 blood group, Cx37- blood group (taken from blood). The expression of Cx37 in different parts of Cx37 rats was detected by RT-PCR. Mononuclear macrophages were isolated from the blood and plaque of Cx37 and Cx37- rats and cultured. The current density was measured by whole-cell recording method and compared. Results the relative expression of Cx37 in mononuclear macrophages in plaque of Cx37 rats was significantly higher than that in blood [(1.1 卤0.02) vs (0.6 卤0.03)], and the difference was statistically significant (P0. 010). Cx37. Cx37- rats were detected carotid plaque by energy dispersive CT. The results showed that the current density of Cx37 plaque group was (0.61 卤0.06), (0.67 卤0.07) at 80120160 ms. Conclusion the effect of Cx37 on the current density in plaque of mononuclear macrophages in atherosclerosis is more significant than that in peripheral blood, which can promote the activation of macrophages and the further development of atherosclerosis.
