腹主动脉瘤组织中HIF-1α、MMP-2及MMP-9的表达及意义

【摘要】:目的探讨低氧诱导因子(HIF)-1α、基质金属蛋白酶(MMP)-2及MMP-9在腹主动脉瘤(AAA)组织中的表达及意义。方法采用免疫组化法检测68例AAA及20例正常腹主动脉组织中HIF-1α、MMP-2及MMP-9的表达,并分析三者与临床病理参数的关系。结果 HIF-1α表达于AAA中膜及外膜平滑肌细胞、巨噬细胞的细胞核中,MMP-2表达于AAA中膜及外膜平滑肌细胞的细胞质中,MMP-9表达于AAA中膜及外膜平滑肌细胞、巨噬细胞的细胞质中,三者在正常腹主动脉组织中均不表达。AAA中HIF-1α、MMP-2及MMP-9的阳性率显著高于正常腹主动脉(P0.001)。AAA中动脉瘤直径≥5.0 cm的患者HIF-1α、MMP-2及MMP-9的表达高于动脉瘤直径5.0 cm者(P0.001);AAA中有发生心脑血管意外史患者HIF-1α、MMP-2及MMP-9的表达高于未发生者(P0.001)。AAA中HIF-1α、MMP-2及MMP-9与年龄、性别、有无吸烟史及高血压无关。同时AAA中HIF-1α、MMP-2及MMP-9表达两两呈正相关。结论 HIF-1α在AAA中高表达,并与MMP-2及MMP-9起协同作用,在AAA的发生发展中起重要作用。
[Abstract]:Objective to investigate the expression and significance of hypoxia inducer (HIF)-1 伪, matrix metalloprotease (MMP)-2 and MMP-9 in (AAA) of abdominal aortic aneurysm. Methods Immunohistochemical method was used to detect the expression of HIF-1 伪, MMP-2 and MMP-9 in 68 cases of AAA and 20 cases of normal abdominal aortic tissue, and the relationship between them and clinicopathological parameters was analyzed. Results HIF-1 伪 was expressed in AAA media and outer membrane smooth muscle cells, MMP-2 was expressed in the nucleus of macrophages, MMP-2 was expressed in the cytoplasm of AAA media and outer membrane smooth muscle cells, and MMP-9 was expressed in AAA media and outer membrane smooth muscle cells. In the cytoplasm of macrophages, none of them were expressed in normal abdominal aortic tissue. HIF-1 伪 in AAA, The positive rate of MMP-2 and MMP-9 was significantly higher than that of normal abdominal aorta (P0.001).
