衰老心肌细胞缺氧后处理中miR-204的作用

【摘要】:目的探索miR-204在衰老心肌细胞缺氧后处理中的作用。方法经D-半乳糖诱导心肌细胞衰老后,随机分为对照组(ONC组)、缺氧复氧组(OAR组)、后处理组(OIPO组)。采用ELISA法检测培养基中c Tn-Ι的含量;微板法检测LDH的含量;XTT法检测细胞活性;荧光定量PCR法检测miR-204的mRNA表达,并确定miR-204与c Tn-Ι、LDH的相关性。结果与ONC组相比,OAR、OIPO组c Tn-Ι、LDH的含量均增加(P0.05,P0.01);与OAR组相比,OIPO组c Tn-Ι、LDH的含量无显著性改变。与ONC组相比,OAR、OIPO组miR-204的表达均降低(P0.01);相关性分析显示miR-204与c Tn-Ι、LDH呈现负相关。结论缺氧后处理对缺氧复氧所致的衰老心肌细胞损伤的保护作用减弱与miR-204有关。
[Abstract]:Objective to explore the role of miR-204 in anoxic post-treatment of aging cardiomyocytes. Methods after cardiomyocyte senescence induced by D-galactose, the cardiomyocytes were randomly divided into control group (ONC group), hypoxia-reoxygenation group (OAR group) and post-treatment group (OIPO group). The content of c Tn- I in culture medium was detected by ELISA method, the content of LDH was detected by microplate method, the cell activity was detected by XTT method, and the mRNA expression of miR-204 was detected by fluorescence quantitative PCR, and the correlation between miR-204 and c Tn- I, LDH was determined. Results compared with ONC group, the contents of c Tn- I and LDH in OAR,OIPO group were increased (P0.05, P 0.01), but the contents of c Tn- I and LDH in OIPO group had no significant change compared with OAR group. Compared with ONC group, the expression of miR-204 in OAR,OIPO group was decreased (P 0.01), and correlation analysis showed that miR-204 was negatively correlated with c Tn- I and LDH. Conclusion the weakening of protective effect of hypoxia post-treatment on aging cardiomyocyte injury induced by hypoxia and reoxygenation is related to miR-204.
