自发性高血压大鼠冠状动脉的收缩功能变化及机制研究

高血压是一种以细小动脉硬化为基本病变的全身性疾病,属于慢性非感染性疾病,它所引起的心、脑和肾等靶器官的损害,其发病率和死亡率也是居高不下,威胁着人类的健康。冠心病是高血压导致的最重要的靶器官损害之一。流行病学调研发现,我国冠心病患者中有一半以上合并高血压,而在正常者中冠心病的患病率比高血压病患者低出4倍。因此,对高血压导致冠状动脉的功能变化及其机制的研究越来越迫在眉睫。目的:研究高血压大鼠冠状动脉收缩和舒张功能变化规律,并探讨其相关的机制。方法:利用SHR和Wistar成年雄性大鼠,周龄为16-18周,采用尾压法检测大鼠的收缩压和舒张压;用小动物B超检测大鼠心脏功能后;采取二氧化碳处死,快速取出心脏,在体视显微镜下分离出大鼠的左右冠状动脉,将部分制备成1.8-2.0 mm长的冠状动脉环,经微血管张力仪测定冠状动脉环的张力,比较不同激动剂诱导高血压冠状动脉收缩的变化,以及运用多种阻断剂干预后,观察其效应的变化,探讨其相关机制。其余部分用于原代培养冠状动脉血管平滑肌细胞(VSMCs),用Fluo4负载后,在激光共聚焦显微镜下检测两组大鼠冠状动脉VSMCs内Ca2+浓度...
