循环microRNA对支架内再狭窄的预测作用及曲美他嗪的预防作用

背景与目的药物洗脱支架(drug-eluting stent, DES)得到广泛应用后,但仍有一定比例的患者发生支架内再狭窄(in-stent restenosis, ISR),在不同时间点识别发生ISR的高危患者具有重要的临床意义。目前能有效预防ISR的药物较少。本课题旨在寻找新的可以预测、筛选ISR患者的方法,同时探讨曲美他嗪(trimetazidine, TMZ)预防ISR的有效性。方法⑴PCI术前循环microRNA预测ISR研究:采用microRNA芯片检测冠心病患者DES植入术前血样,比较30例ISR患者和30例非ISR(NISR)患者术前循环血中microRNA的表达差异,定量RT-PCR(qRT-PCR)验证芯片结果得到候选microRNA。在另一组患者(ISR患者及NISR患者各178例)中用qRT-PCR检测每例患者候选microRNA的表达水平,Logistic回归分析得到与ISR相关的目的microRNA,数据转换建立microRNA积分,ROC法分析该积分对ISR的预测作用。⑵随访期间循环microRNA筛选ISR研究:采用microRNA芯片检测复查造影术前...
