CD147、基质金属蛋白酶-2和血管内皮生长因子在氧诱导视网膜病变大鼠中的表达

【摘要】:目的建立氧诱导视网膜病变大鼠模型,观察其视网膜各层组织中CD147、基质金属蛋白酶(MMP)-2和血管内皮生长因子(VEGF)的表达情况。方法1、将68只新生Wistar大鼠随机分为高氧组和对照组。每组各34只大鼠。高氧诱导实验组形成氧诱导视网膜病变模型;对照组不予任何处理。2、新生鼠7、14日龄时,取出眼球后行视网膜铺片,观察视网膜血管形态学变化。3、新生鼠14日龄时,取出眼球后行视网膜石蜡切片,计数突破视网膜内界膜的内皮细胞核数。4、新生鼠12、14、16日龄时,取眼球行免疫组织化学染色法,检测视网膜各层组织中CD147、MMP-2和VEGF的表达。5、运用双因素相关分析法,解析氧诱导视网膜病变模型中CD147表达与MMP-2、VEGF表达的关联性。结果1、新生鼠7日龄时,对照组大鼠视网膜血管自视盘呈放射状散开,走形清晰,血管径较粗;高氧组大鼠视网膜血管收缩、弯曲,周边散在广泛的无血管区,或仅见为数不多的小面积的血管组织网。新生鼠14日龄时,对照组大鼠视网膜血管形态发育较好,走形正常,网格规整;高氧组大鼠出现视网膜血管增生、纡曲、扩张,并于灌注区与非灌注区交界处形成新生血管丛,部分发生渗出、出血。2、新生鼠14日龄时,对照组、高氧组突破网膜内界膜的内皮细胞核数分别为(1.30±1.26)、(19.70±3.56)个;两组比较,差异有统计学意义(t=21.813,P0.01)。3、免疫组织化学染色结果显示,对照组大鼠视网膜各层均可见CD147、MMP-2及VEGF有少量表达。而高氧组大鼠视网膜各层均可见CD147、MMP-2、VEGF有显著表达。在新生鼠12、14、16日龄时,将两组大鼠视网膜各层中CD147(t=5.612、 11.390、6.355)、MMP-2(t=4.122、8.047、4.422)、VEGF (t=4.955、12.176、5.110)表达相较,差异均有统计学意义(P0.01)。4、采用相关性分析发现,CD147的表达与MMP-2、VEGF的表达均具有正相关性(r=0.755、0.820,P0.01)。结论CD147、MMP-2、VEGF在氧诱导视网膜病变大鼠视网膜中有大量表达。
[Abstract]:Objective to establish a rat model of oxygen-induced retinopathy. To observe the expression of CD147, matrix metalloproteinase-2 (MMP-2) and vascular endothelial growth factor (VEGF) in various layers of retina. Methods 1. Sixty-eight newborn Wistar rats were randomly divided into hyperoxia group and control group, 34 rats in each group were induced by hyperoxia. Oxygen induced retinopathy model was formed in the experimental group. In the control group, no treatment was given. At the age of 714 days, retinal slices were taken out and retinal vascular morphology was observed. At the age of 14 days, paraffin sections of the retina were taken out from the eyes of the newborn rats. The number of endothelial nuclei that broke through the inner membrane of the retina was counted. At the age of 1416 days, the eyes of the newborn rats were taken for immunohistochemical staining to detect the expression of CD147 MMP 2 and VEGF in all layers of the retina, and the expression of MMP-2 and VEGF were detected by double factor correlation analysis. Results 1. At the age of 7 days, the retinal vascular self-vision disc of the control group was radially dispersed, the shape was clear, and the diameter of the blood vessel was relatively large. The retinal vessels in hyperoxia group were constricted, bent and scattered in a wide, non-vascular area, or only a few small vascular tissue networks were found. At 14 days of age, the retinal vessels in the control group developed well and the shape of retinal vessels was normal. In hyperoxia group, retinal vessels proliferated, tortuous, dilated, and formed neovascularization plexus at the junction of perfusion area and non-perfusion area. The number of endothelial nuclei in the hyperoxia group was 1.30 卤1.26, 19.70 卤3.56, respectively, and the difference was statistically significant between the two groups. The results of immunohistochemical staining showed that the number of endothelial nuclei in the hyperoxia group was 21.813 (P 0.01), and that in the hyperoxia group was higher than that in the control group (P < 0.05). The expression of CD147MMP 2 and VEGF were observed in all layers of the retina of rats in the control group, while in the hyperoxia group, the expression of CD147MMP 2 was found in all layers of the retina of the hyperoxia group. At the age of 1416 days, the expression of CD147MMP 2 in the different layers of the retina of the two groups was 5.612, 11.3906.355MMP 2t was 4.1228.047.4.422VEGF-VEGFtt 4.955n12.1765.110), and the expression of CD147MMP 2 in the different layers of the retina of the control group was higher than that of the control group. The correlation analysis showed that there was a positive correlation between the expression of CD147 and the expression of MMP-2VEGF. Conclusion the expression of CD147 and MMP-2VEGF is highly expressed in oxygen-induced retinopathy rats.
