12-脂氧化酶与糖尿病性视网膜病变发生发展相关性的研究

【摘要】: 目的:通过对比糖尿病性视网膜病变(DR)大鼠视网膜内12-脂氧化酶的表达与正常大鼠的差异,证明12-脂氧化酶与糖尿病性视网膜病变发生发展的相关性。为进一步研究DR的治疗方法打下理论基础。 方法:建立链脲佐菌素(STZ)诱导糖尿病性视网膜病变大鼠模型,监测血糖和体重变化情况,5个月后观察实验组和对照组大鼠眼球形态学变化,采用RT-PCR的方法测定大鼠视网膜内12-LOmRNA表达情况。 结果:实验组分为糖尿病大鼠血糖高组和糖尿病大鼠血糖恢复组。糖尿病大鼠血糖高组并发性白内障发生率达100%,部分大鼠出现虹膜新生血管、周边视网膜及视盘前新生血管。正常组大鼠视网膜12-LOmRNA相对表达量为1.17±0.18,糖尿病大鼠血糖高组为1.96±0.28,糖尿病大鼠血糖恢复组为1.64±0.33;实验组与正常组比较表达量明显升高,差异具有显著性意义,糖尿病大鼠血糖恢复组与糖尿病大鼠血糖高组比较略有减少,但差异不明显。 结论:链脲佐菌素可以有效诱导大鼠糖尿病性白内障及糖尿病性视网膜病变,造模5个月大鼠可产生增殖期DR改变。STZ诱导的DR大鼠12-LO表达明显升高,且血糖恢复正常对表达无明显影响。
[Abstract]:Objective: to compare the expression of 12-lipoxygenase in the retina of diabetic retinopathy rats and normal rats. To prove the correlation between 12-lipoxygenase and the occurrence and development of diabetic retinopathy, and to lay a theoretical foundation for the further study of the treatment of Dr. Methods: the diabetic retinopathy model was induced by streptozotocin (STZ), and the changes of blood glucose and body weight were monitored. After 5 months, the morphologic changes of the eyes of the experimental group and the control group were observed. The expression of 12-LOmRNA in rat retina was determined by RT-PCR. Results: the experimental group was divided into two groups: diabetic rats with hyperglycemia and diabetic rats with recovery of blood glucose. The incidence of cataract in diabetic rats was 100%, and iris neovascularization was found in some rats. The relative expression of 12-LOmRNA was 1.17 卤0.18 in the normal group, 1.96 卤0.28 in the hyperglycemia group and 1.64 卤0.33 in the diabetic recovery group. There was significant difference between diabetic rats and diabetic rats, but there was no significant difference between diabetic rats and diabetic rats. Conclusion: streptozotocin can effectively induce diabetic cataract and diabetic retinopathy in rats. The 12-LO expression of Dr rats induced by streptozotocin can be significantly increased after 5 months of modeling. And there was no obvious effect on the expression of blood glucose returning to normal.
