BUB1B mRNA在喉鳞状细胞癌中的表达及意义

【摘要】: 目的 通过检测BUB1B mRNA在喉鳞状细胞癌及其相邻正常喉黏膜组织中的表达,探讨BUB1B基因在喉鳞状细胞癌发生发展中的作用及其临床意义。 材料与方法 一、标本的来源与制备 所选30例喉鳞癌标本均取自中国医科大学附属第一医院耳鼻咽喉科2009年11月至2010年2月手术切除标本,所有患者术前均未行放疗和化疗,均无远隔器官转移,术后病理示其切缘皆为阴性。每例分别取新鲜癌组织及癌旁1 cm以外非癌喉黏膜组织-70℃冻存备用。 二、实验方法 提取总RNA,反转录合成cDNA,以β-actin为内参照,应用逆转录-聚合酶链反应方法检测30例喉鳞状细胞癌组织及相邻癌旁正常黏膜组织中BUBIB mRNA表达情况。 结果 以BUBIB和β-act in的光密度值之比做为BUB1B mRNA表达的相对值,半定量分析BUB1B mRNA在喉鳞状细胞癌组织和相邻正常喉黏膜组织中的表达。喉鳞状细胞癌组织中BUBIB/β-actin为0.215±0.076,癌旁正常喉黏膜组织为0.317±0.128,经统计学分析(配对t检验),2组数据差异具有统计学意义,(t=4.521,P0.05),采用独立样本t检验的方法,对其在临床分型、组织学分组及淋巴结转移等方面进行统计学分析,其表达在不同临床分期中有差异,而在不同临床分型、组织学分级及淋巴结转移转移中无明显差异(P>O.05)。 BUB1B mRNA在喉癌组织中低表达,且其低表达可能与喉癌的发生发展有关。
[Abstract]:objective
By detecting the expression of BUB1B mRNA in laryngeal squamous cell carcinoma and its adjacent normal laryngeal mucosa, the role of BUB1B gene in the development of laryngeal squamous cell carcinoma and its clinical significance were investigated.
Materials and methods
First, the source and preparation of the specimen
All the 30 specimens of laryngeal squamous cell carcinoma were taken from the surgical excision specimens from the Department of Otolaryngology, the First Affiliated Hospital of China Medical University, from November 2009 to February 2010. None of the patients were treated with radiotherapy and chemotherapy before operation. No distant organ metastasis was performed. The membrane tissue is stored at -70 C for storage.
Two, experimental method
The total RNA, reverse transcriptional synthesis of cDNA and beta -actin as internal reference were used to detect the expression of BUBIB mRNA in 30 cases of laryngeal squamous cell carcinoma and adjacent normal mucosa tissues by reverse transcription polymerase chain reaction.
Result
Using the ratio of BUBIB and beta -act in as the relative value of BUB1B mRNA expression, the expression of BUB1B mRNA in the tissues of laryngeal squamous cell carcinoma and adjacent normal laryngeal mucosa was semi quantified. The BUBIB/ beta -actin in the squamous cell carcinoma of the larynx was 0.215 + 0.076, and the normal laryngeal mucosa adjacent to the carcinoma was 0.317 + 0.128, and was statistically analyzed (paired t). The difference between the 2 groups was statistically significant, (t=4.521, P0.05). The methods of independent sample t test were used to analyze the clinical classification, histologic grouping and lymph node metastasis, and the expression was different in different clinical stages, but in different clinical classification, histology classification and lymph node metastasis. Significant differences (P > O.05).
The expression of BUB1B mRNA is low in laryngeal carcinoma, and its low expression may be related to the development of laryngeal carcinoma.
