脂质体槲皮素对DM大鼠视网膜糖基化终产物生成及受体表达的影响

【摘要】: 目的观察脂质体槲皮素(Liposomal Quercertin, LQ)对糖尿病(diabetes mellitus,DM)大鼠视网膜糖基化终产物(advanced glycation end products, AGEs)生成及受体(RAGE mRNA)表达的影响,并探讨可能的作用机制。 方法采用蒸发-高压均质法制备脂质体槲皮素悬浊液,链脲佐菌素(streptozocin,STZ)法诱导DM大鼠模型,并将DM大鼠随机分为DM模型组、脂质体槲皮素低、中、高剂量灌胃组(50mg/kg.d、150mg/kg.d及250mg/kg.d)及氨基胍灌胃组(100mg/kg.d),连续给药12周;实验结束时,HE染色观察视网膜形态学变化,免疫荧光法、RT-PCR方法分别检测DM大鼠视网膜中AGEs的沉积情况及RAGE mRNA的表达,并与正常对照组比较。 结果 1.视网膜形态学变化:DM模型组视网膜各层出现典型的DM病理学改变,脂质体槲皮素中剂量组视网膜病理学改变较DM模型组减轻。 2.脂质体槲皮素各剂量组大鼠体重较DM模型组无明显变化,血糖较DM模型组略有下降,但差异无统计学意义。 3.较DM模型组结果(AGEs:0.922±0.005、RAGE mRNA:1.192±0.098)比较,脂质体槲皮素各组及氨基胍组视网膜中AGEs沉积减少,RAGEmRNA表达也降低(P0.05),以脂质体槲皮素中剂量组(AGEs:0.734±0.007、RAGE mRNA:0.724±0.028)降低最为明显(P0.01)。 结论 1.脂质体槲皮素能够在一定程度上减轻视网膜形态学病理改变,改善STZ性DM大鼠一般状况,可能有降血糖的作用,尚需进一步实验证实。 2.脂质体槲皮素可通过有效减少DM大鼠视网膜中AGEs的沉积,下调RAGE mRNA的表达,即通过抑制非酶糖基化的形成,早期干预视网膜微血管病变的发生。 3.本研究为脂质体槲皮素的下一步临床应用研究打下基础,同时也对探索DM的临床中药治疗提供新的思路和理论依据。
[Abstract]:Objective to investigate the effects of liposomal Quercerin (LQQ) on the production of advanced glycation end products, AGEs) and the expression of receptor rage mRNAs in retina of diabetic rats with diabetes mellitus Mellitus (DMN), and to explore the possible mechanism. Methods Liposome quercetin suspension was prepared by evaporation-high pressure homogenization method, streptozocin STZ (streptozocin STZ) method was used to induce DM rat model, and DM rats were randomly divided into DM model group with low and moderate quercetin liposome. 50 mg / kg 路d and 250 mg / kg 路d in high dose gavage group and 100 mg / kg 路d ~ (-1) aminoguanidine group respectively for 12 weeks. At the end of the experiment, morphological changes of retina were observed by HE staining, and AGEs deposition and RAGE mRNA expression in retina of DM rats were detected by immunofluorescence RT-PCR. And compared with the normal control group. Result 1. The pathological changes of retina in the middle dose of liposome quercetin group were less than those in the DM model group. 2. There was no significant change in body weight and blood glucose in each dose of liposome quercetin group as compared with DM group, but the difference was not statistically significant. 3. Compared with DM model group, ages: 0.922 卤0.005 rage mRNA:1.192 卤0.098), the deposition of AGEs in retina of quercetin group and aminoguanidine group was also decreased (P 0.05), especially in the middle dose group of liposome quercetin 0.734 卤0.007 rage mRNA:0.724 卤0.028). Conclusion 1. The liposome quercetin can alleviate the pathological changes of retinal morphology and improve the general condition of STZ diabetic rats to a certain extent, which may have the effect of hypoglycemia, which needs further experimental verification. 2. Liposome quercetin can effectively reduce the deposition of AGEs and down-regulate the expression of RAGE mRNA in the retina of DM rats, that is, by inhibiting the formation of non-enzymatic glycosylation, early intervention of retinal microangiopathy occurs. 3. This study laid a foundation for the further clinical application of liposome quercetin and provided new ideas and theoretical basis for exploring the clinical treatment of DM.
