人晶状体上皮细胞中bax的表达与白内障形成的相关性研究

【摘要】: 目的:本研究应用免疫组织化学SP法染色,检测凋亡相关基因bax在年青人白内障患者(年龄在40-55岁之间)晶状体上皮细胞中的表达及表达程度,以探讨晶状体上皮细胞的凋亡与年青人白内障发生的关系。 方法:采用链霉菌亲生物素蛋白—过氧化物酶标免疫组织化学法(steptavidin-peroxidase,SP)染色,光学显微镜下检测18例年青人白内障患者(实验组)和22例老年性白内障患者(对照组)的晶状体上皮细胞的bax基因的表达及表达程度。 结果:在低龄组年青人白内障患者的晶状体前囊膜上皮细胞胞浆中可见棕黄色颗粒均匀分布,提示bax基因在年青人白内障患者的晶状体上皮细胞中有表达,其表达程度:bax基因在18例年青人白内障患者的晶状体上皮细胞中强阳性表达10例,阳性表达6例,弱阳性表达2例,无阴性表达病例;在高龄组老年性白内障患者的晶状体前囊膜上皮细胞胞浆中可见棕黄色颗粒均匀分布,提示bax基因在高龄老年性白内障患者晶状体上皮细胞中有表达,其表达程度:bax基因在22例高龄白内障患者晶状体上皮细胞中强阳性表达12例,阳性表达7例,弱阳性表达3例,无阴性表达病例。凋亡相关基因bax在低龄和高龄组白内障患者的晶状体上皮细胞中均有明显的表达,且两组间bax基因的表达没有显著性差异(P0.05)。 结论:年青人白内障(40-55岁)与老年性白内障(60-80岁)发病的最初诱因可能不同,但最终都因晶状体上皮细胞的凋亡而导致白内障的发生。
[Abstract]:Objective: to detect the expression and expression of apoptosis-related gene bax in lens epithelial cells of young cataract patients (aged 40-55 years) by immunohistochemical SP staining. To investigate the relationship between lens epithelial cell apoptosis and cataract in young people. Methods: Streptomyces biophile protein and peroxidase labeled immunohistochemical method was used for the staining of Streptomyces vidin-peroxidase (SPX). The expression and degree of bax gene in lens epithelial cells of 18 young cataract patients (experimental group) and 22 senile cataract patients (control group) were detected under optical microscope. Results: there were brown granules in the cytoplasm of lens anterior capsule epithelial cells in young cataract patients, suggesting that bax gene was expressed in lens epithelial cells of young cataract patients. There were 10 cases of strong positive expression, 6 cases of positive expression, 2 cases of weak positive expression, and no case of negative expression. The distribution of brown granules in the cytoplasm of lens anterior capsule epithelial cells in the elderly patients with senile cataract showed that bax gene was expressed in lens epithelial cells of senile cataract patients. There were 12 cases of strong positive expression, 7 cases of positive expression, 3 cases of weak positive expression and no negative expression in 22 cases of senile cataract. The expression of apoptosis-related gene bax in lens epithelial cells of both young and aged cataract patients was significant, and there was no significant difference in the expression of bax gene between the two groups (P 0.05). Conclusion: the initial inducement of cataract in young people aged 40-55 years may be different from that in senile cataract patients (60-80 years old), but eventually it is caused by the apoptosis of lens epithelial cells.
