Ruboxistaurin对小鼠氧诱导视网膜病变中视网膜VEGF表达的影响

【摘要】: 目的:通过建立氧诱导视网膜病变(OIR)小鼠模型模拟人类未成熟儿视网膜病变。予以PKC-β抑制剂Ruboxistaurin灌胃,检测Ruboxistaurin对视网膜组织中VEGF表达的变化情况。 方法:建立氧诱导视网膜病变模型。随机对120只7天龄C57BL/6J幼鼠分为正常对照组、高氧模型组、药物治疗组和空白对照组,除正常对照组外,其余幼鼠均暴露在浓度为(75±2)%的高氧状态下5天,随后正常氧环境下5天,建成氧诱导视网膜病变模型。药物治疗组和空白对照组又分别按照灌胃剂量的不同分为大,中,小剂量三个亚组。用免疫组织化学染色与逆转录—聚合酶链反应法(RT-PCR)检测视网膜组织中VEGF含量的变化。 结果:免疫组织化学染色显示:正常对照组中,VEGF在视网膜组织中即有少量表达;高氧模型组中其表达水平升高,与正常对照组比较差异有统计学意义(P0.01), RT-PCR产物凝胶电泳检测VEGFmRNA的含量与免疫组织化学结果一致。药物治疗组的三个亚组VEGF蛋白灰度值分别为132.7713±5.5100,138.0096±1.8700,143.2910±4.6600, VEGFmRNA相对表达量分别为0.8973±0.0600,0.8254±0.0400,0.6183±0.0300,正常对照组、高氧模型组,药物治疗组各组比较差异均具有统计学意义(P0.05),高氧模型组和空白对照组比较差异无统计学意义(P0.05) 结论:VEGF在氧诱导视网膜病变小鼠模型的视网膜中表达增强。OIR模型的视网膜组织中VEGF表达部分是通过PKC通路产生的。Ruboxistaurin可以减少OIR模型鼠视网膜组织中的VEGF的表达。
[Abstract]:Aim: to establish oxygen-induced retinopathy (OIR) mouse model to simulate human immature retinopathy. PKC- 尾 inhibitor Ruboxistaurin was administered intragastrically to detect the changes of VEGF expression in retina. Methods: oxygen-induced retinopathy model was established. 120 7-day-old C57BL/6J young rats were randomly divided into normal control group, hyperoxia model group, drug treatment group and blank control group. Except for normal control group, the other young rats were exposed to hyperoxia (75 卤2%) for 5 days and then to normal oxygen environment for 5 days. Oxygen-induced retinopathy model was established. The drug treatment group and the blank control group were divided into three subgroups: large, middle and low dose according to the different dosage of gastric perfusion. Immunohistochemical staining and reverse transcriptase polymerase chain reaction (RT PCR) were used to detect the content of VEGF in retina. Results: immunohistochemical staining showed that there was a little expression of VEGF in the retina of the normal control group, but the expression level of VEGF in the hyperoxia model group was higher than that in the hyperoxia model group. Compared with the control group, the difference was statistically significant (P 0.01). The content of VEGFmRNA detected by gel electrophoresis of RT-PCR products was consistent with the results of immunohistochemistry. The gray values of VEGF protein in the three subgroups of the drug treatment group were 132.7713 卤5.100 138.0096 卤1.8700143.2910 卤4.66000.The relative expression of VEGFmRNA was 0.8973 卤0.0600,0.8254 卤0.0400,0.6183 卤0.0300, respectively. There was no significant difference between hyperoxia model group and blank control group (P 0.05). Conclusion the expression of VEGF in the retina of mice with oxygen-induced retinopathy is enhanced. The expression of VEGF in the retina of the mouse model of OIR induced retinopathy is partly induced by the PKC pathway. Ruboxistaurin can reduce the expression of VEGF in the retina of OIR model mice.
