RECK、RAGE和MMP-9基因在鼻咽癌中的表达及意义

【摘要】: 目的检测永生化鼻咽上皮细胞系NP69-SV40T和具有不同转移倾向的鼻咽癌细胞系SUNE-1亚株-6-10B(只成瘤,不转移)、SUNE-1亚株-5-8F(高成瘤,高转移)中,伴有kazal域的富含半胱氨酸的逆转诱导蛋白(reversion-inducing-cysteine-rich protein with kazal motifs,RECK)、高级糖基化终末产物受体(receptors for advanced glycation end products)及基质金属蛋白酶9(matrix metalloproteinase 9,MMP-9)的mRNA表达情况。 方法应用实时荧光定量PCR技术检测永生化鼻咽上皮细胞系NP69-SV40T、鼻咽癌细胞系SUNE-1亚株-6-10B、SUNE-1亚株-5-8F中RECK、RAGE及MMP-9 mRNA的表达。 结果MMP-9 mRNA在鼻咽癌细胞系中的表达较永生化鼻咽上皮细胞系NP69-SV40T下调(P0.05),鼻咽癌细胞系5-8F组表达稍高于6-10B组,两者间差异无显著性(P0.05);RAGE mRNA在鼻咽癌细胞系中的表达较永生化鼻咽上皮细胞系NP69-SV40T稍下调,三种细胞系中的表达差异无显著性(P0.05);RECK mRNA在三种细胞系中的表达差异亦无显著性(P0.05)。 结论体外条件下RAGE、RECK和MMP-9 mRNA的表达水平与细胞系性质无明显关联,与NPC细胞系潜在的转移能力无明显关联,三者之间亦无显著相关。 目的检测伴有kazal域的富含半胱氨酸的逆转诱导蛋白(reversion-inducing-cysteine-rich protein with kazal motifs, RECK)、高级糖基化终末产物受体(receptors for advanced glycation end products, RAGE)及基质金属蛋白酶9(matrix metalloproteinase 9, MMP-9)在鼻咽癌(nasopharyngeal carcinoma, NPC)组织中的表达情况及其相关性。 方法运用免疫组化方法检测64例NPC组织(伴颈淋巴结转移者30例,无颈淋巴结转移者34例)和30例慢性鼻咽炎组织(chronic nasopharyngitis tissue, CNT)中RECK、RAGE和MMP-9蛋白的表达。 结果RECK在CNT及NPC组织表达阳性率极低,且两者间差异无显著性(P 0.05),但周边炎症细胞及癌巢周边基质中RECK强表达,在伴与不伴颈淋巴结转移组中的阳性率分别为3.3%(1/30)和11.8%(4/30),两者间差异无显著性(P 0.05);RAGE表达阳性率在CNT和NPC组织分别为100% (30/30)和70.3% (45/64),MMP-9则分别为46.7%(14/30)和78.1% (50/64);RAGE在伴与不伴颈淋巴结转移组中的阳性率分别为86.7%(26/30)和55.9%(19/34),MMP-9则分别为90.0%(27/30)和67.7%(23/34),各组间差异均具统计学意义(P0.05)。在NPC组织中,RECK与MMP-9的表达呈负相关(r=-0.369,p0.01), RAGE与MMP-9则呈正相关(r=0.471,p0.01)。 结论RECK、RAGE在NPC组织较CNT中表达下调,MMP-9则上调;在伴颈淋巴结转移的NPC组织中,RECK表达下调,RAGE、MMP-9的表达上调。三者的异常表达可能与NPC的进展有关,RECK、RAGE可能通过调节MMP-9的表达参与NPC的侵袭转移过程。
[Abstract]:Objective to detect the relationship between immortalized nasopharyngeal epithelial cell line NP69-SV40T and nasopharyngeal carcinoma cell line SUNE-1 substrain -6-10B (tumorigenic, non-metastatic) substrain -5-8F (Gao Cheng tumor, high metastasis). MRNA expression of cysteine-rich reverse inducible protein (reversion-inducing-cysteine-rich protein with kazal motifs), advanced glycosylation end product receptor (receptors for advanced glycation end products) and matrix metalloproteinase-9 (matrix metalloproteinase 9 (MMP-9) with kazal domain. Methods Real-time quantitative PCR was used to detect the mRNA expression of RECKnRAGE and MMP-9 in immortalized nasopharyngeal epithelial cell line NP69-SV40T and nasopharyngeal carcinoma cell line SUNE-1 substrain -6-10BU SUNE-1 substrain -5-8F. Results the expression of MMP-9 mRNA in nasopharyngeal carcinoma cell line was lower than that in immortalized nasopharyngeal epithelial cell line NP69-SV40T (P0.05). The expression of MMP-9 mRNA in nasopharyngeal carcinoma cell line 5-8F was slightly higher than that in 6-10B group. The expression of rage mRNA in nasopharyngeal carcinoma cell line was lower than that in immortalized nasopharyngeal epithelial cell line NP69-SV40T (P0.05). Conclusion there is no significant correlation between the expression level of rack and MMP-9 mRNA in vitro and the nature of the cell line, and the potential metastasis ability of NPC cell line, and there is no significant correlation between them. Objective to investigate the expression of cysteine-rich reverse inducible protein (reversion-inducing-cysteine-rich protein with kazal motifs, reck), advanced glycation end product receptor (receptors for advanced glycation end products, rage) and matrix metalloproteinase-9 (MMP-9) in nasopharyngeal carcinoma (NPC) tissues with kazal domain. Reach the situation and its relevance. Methods Immunohistochemical method was used to detect the expression of RECK rage and MMP-9 protein in 64 NPC tissues (30 cases with cervical lymph node metastasis, 34 cases without cervical lymph node metastasis) and 30 cases with chronic nasopharyngitis. Results the positive rate of reck in CNT and NPC tissues was very low, and there was no significant difference between them (P 0.05). The positive rates of rage were 3.3% (1 / 30) and 11.8% (4 / 30) in patients with and without cervical lymph node metastasis, respectively. There was no significant difference between them (P 0.05). The positive rates of rage expression in CNT and NPC tissues were 100% (30 / 30) and 70.3% (45 / 64), respectively, 46.7% (14 / 30) and 78.1% (50 / 64) in CNT and NPC tissues. The positive rates of MMP-9 in metastatic group were 86.7% (26 / 30) and 55.9% (19 / 34), respectively. The positive rates of MMP-9 in metastatic group were 90.0% (27 / 30) and 67.7% (23 / 34), respectively (P0.05). In NPC tissues, the expression of reck was negatively correlated with MMP-9 (r-0.369, p0.01), while rage was positively correlated with MMP-9 (r-0.471p0.01). Conclusion the down-regulated expression of MMP-9 was up-regulated in NPC tissues compared with CNT, and down-regulated in NPC with cervical lymph node metastasis, and the expression of RAGE-MMP-9 was up-regulated in NPC tissues with cervical lymph node metastasis. The abnormal expression of these three proteins may be related to the progression of NPC, which may be involved in the invasion and metastasis of NPC by regulating the expression of MMP-9.
