FoxM1在喉癌中的表达及其在姜黄素抑制喉癌细胞株Hep-2增殖、侵袭、转移中的作用

【摘要】:目的: 1、研究FoxM1在喉癌中的表达情况及临床意义,为喉癌的发生发展、预后评估及寻求新的治疗靶点提供实验依据。 2、研究姜黄素对喉癌细胞株Hep-2增殖、侵袭、转移能力的影响,为喉癌治疗提供新的策略。 3、研究FoxM1在姜黄素作用喉癌细胞株Hep-2前后的表达变化,探讨FoxM1在其中的作用。 方法: 1、运用免疫组织化学方法检测89例喉癌及89例癌旁组织、20例喉乳头状瘤组织中FoxM1蛋白的表达情况,分析它们在不同病变组织中的表达情况及与喉癌临床病理参数的关系。 2、运用四甲基偶氮唑蓝(MTT)法、侵袭实验以及迁移实验检测经姜黄素作用后喉癌细胞Hep-2体外增殖、侵袭和转移能力的变化。 3、运用反转录-聚合酶链反应(RT-PCR)检测姜黄素作用前后喉癌细胞中FoxM1的表达水平。 结果: 1、FoxM1蛋白表达的阳性率在喉癌、喉乳头状瘤及癌旁组织中逐渐降低,其差异有统计学意义(P0.05);FoxM1蛋白的表达随组织分化程度增高而表达降低(P0.05);随T分期增高而表达增加(P0.05);有淋巴结转移组高于无淋巴结转移组(P0.05),随临床分期的增加而表达增加(P0.05)。 2、姜黄素能抑制喉癌细胞Hep-2体外增殖能力,并随着姜黄素浓度的增加和作用时间的延长,其对Hep-2细胞增殖抑制率也增加。另外,姜黄素也抑制Hep-2细胞的侵袭和转移能力,随着药物浓度的增加,其抑制作用也增强。 3、经姜黄素作用后Hep-2细胞中FoxM1mRNA水平较作用前明显下降,差异有统计学意义(P0.05)。 结论: 1、在喉癌中FoxM1蛋白表达水平的增加可能与喉癌的发生、浸润转移密切相关。 2、姜黄素能抑制喉癌细胞的增殖、侵袭、转移能力,其中的机制可能与抑制FoxM1表达相关。
[Abstract]:Objective: 1. To study the expression and clinical significance of FoxM1 in laryngeal carcinoma. To study the effect of curcumin on the proliferation, invasion and metastasis of laryngeal carcinoma cell line Hep-2. To provide a new strategy for the treatment of laryngeal cancer. 3. To study the expression changes of FoxM1 before and after curcumin treatment in laryngeal carcinoma cell line Hep-2, and to explore the role of FoxM1 in the treatment of laryngeal carcinoma cell line Hep-2. Methods: 1. The expression of FoxM1 protein in 89 cases of laryngeal carcinoma and 89 cases of adjacent laryngeal papilloma was detected by immunohistochemical method. Their expression in different pathological tissues and their relationship with the clinicopathological parameters of laryngeal carcinoma were analyzed. 2. MTT assay was used. Invasion assay and migration assay were used to detect the proliferation of laryngeal carcinoma cell line Hep-2 after curcumin treatment in vitro. 3. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of FoxM1 in laryngeal cancer cells before and after curcumin treatment. Results: 1 the positive rate of FoxM1 protein expression in laryngeal carcinoma, laryngeal papilloma and adjacent tissues decreased gradually. The difference was statistically significant (P0.05). The expression of FoxM1 protein decreased with the increase of tissue differentiation (P0.05), increased with the increase of T stage (P0.05). The expression of FoxM1 protein in the group with lymph node metastasis was higher than that in the group without lymph node metastasis (P0.05), and it was expressed with the increase of clinical stage. 2. Curcumin could inhibit the proliferation of laryngeal carcinoma cell line Hep-2 in vitro. The inhibitory rate of curcumin on Hep-2 cell proliferation was also increased with the increase of curcumin concentration and time. In addition, curcumin also inhibited the invasion and metastasis of Hep-2 cells. With the increase of drug concentration, the inhibitory effect of curcumin on Hep-2 cells was also enhanced. The expression of FoxM1 mRNA in Hep-2 cells decreased significantly after curcumin treatment. The difference was statistically significant (P0.05). Conclusion: 1. The increase of FoxM1 protein expression in laryngeal carcinoma may be closely related to the occurrence, invasion and metastasis of laryngeal carcinoma. Curcumin can inhibit the proliferation, invasion and metastasis of laryngeal carcinoma cells. The mechanism may be related to the inhibition of FoxM1 expression.
