SHIP2在喉鳞癌中的表达及意义

【摘要】: 目的:检测SHIP2蛋白在喉鳞状细胞癌(喉鳞癌)组织及其细胞系中的表达,并分析其表达与喉鳞癌临床病理特征及预后之间的关系。 方法:1).运用Western blot技术检测SHIP2蛋白在喉鳞癌细胞株Hep-2的表达情况;2).采用免疫组织化学技术在54例喉鳞癌及16例癌旁粘膜石蜡组织标本中检测SHIP2蛋白的表达水平,并统计分析SHIP2表达水平与喉鳞癌临床病理特征及预后的关系。 结果:SHIP2蛋白在喉鳞癌细胞株Hep-2中的表达较上呼吸道永生化粘膜上皮细胞株NP-69显著上调(0.88±0.09VS0.17±0.07);在喉鳞癌组织中的表达较癌旁粘膜组织的阳性率亦显著增高(77.78%VS18.75%);且其表达水平与喉鳞癌的T分期(T1+T2/T3+T4) (x2=10.119,P=0.006)、临床分期(Ⅰ期+Ⅱ期/Ⅲ期+Ⅳ期)(x2=13.460,P=0.001)、转移(x2=12.959,P=0.002)及复发(x2=11.241,P=0.004)密切相关;SHIP2高表达患者较低表达患者复发快(中位复发时间29.346月±3.605月VS51.321月±2.975月,x2=16.336,P0.001),中位生存时间(41.231月±3.164月)亦显著短于SHIP2低表达患者(56.036月±2.267月)(x2=19.893,P0.001);且SHIP2表达水平结合临床分期、淋巴结有无转移对预后具有更好的预测作用。多因素Cox比例风险回归模型结果显示,SHIP2表达水平与淋巴结有无转移同为喉鳞癌预后的独立影响因素。 结论:本研究发现SHIP2蛋白在喉鳞癌组织及细胞中的表达显著升高,并与喉鳞癌的复发、转移及预后密切相关。这些结果均提示SHIP2可能在喉鳞癌的发生、发展中发挥了重要作用,有望成为预测喉鳞癌复发、转移及预后的重要分子标志物及潜在的治疗靶点。
[Abstract]:Aim: to detect the expression of SHIP2 protein in laryngeal squamous cell carcinoma (LSCC) tissues and its cell lines, and to analyze the relationship between SHIP2 expression and clinicopathological features and prognosis of laryngeal squamous cell carcinoma (LSCC). Method 1). The expression of SHIP2 protein in laryngeal squamous cell carcinoma cell line Hep-2 was detected by Western blot. The expression of SHIP2 protein was detected by immunohistochemistry in 54 cases of laryngeal squamous cell carcinoma and 16 cases of paraffin tissue of adjacent mucosa. The relationship between the expression level of SHIP2 and clinicopathological features and prognosis of laryngeal squamous cell carcinoma was analyzed statistically.
