αB-晶状体蛋白对大鼠急性高眼压后视网膜神经节细胞存活及凋亡蛋白Caspase-3表达的影响

【摘要】:目的 观察αB-晶状体蛋白对大鼠急性高眼压后视网膜神经节细胞存活及凋亡蛋白Caspase-3表达的影响。 方法 3月龄雄性Sprague-Dawley(SD)大鼠,体重250±20g ,160只完全随机设计分成以下4组:A组(急性高眼压组),B组(急性高眼压+玻璃体穿刺组),C组(急性高眼压+玻璃体注射生理盐水组),D组(急性高眼压+玻璃体注射αB-晶状体蛋白组),应用荧光金逆行标记计数法、免疫荧光法观察急性高眼压后各组大鼠视网膜神经节细胞在不同时间点的存活率及凋亡蛋白Caspase-3表达情况。 结果 急性高眼压后实验动物右眼(实验眼)视网膜神经节细胞开始减少,低倍荧光显微镜下可见全视网膜节细胞均在减少,第一周减少最为明显,两周后减少放缓。与其它3组相比,D组在急性高眼压后1周、2周、3周、4周视网膜神经节细胞存活率明显提高,差异具有统计学意义(P0.01),其它3组之间,差异无统计学意义(P0.05);急性高眼压后实验动物右眼(实验眼)Caspase-3免疫荧光染色出现阳性表达,与其它3组相比,D组在急性高眼压后24小时、48小时、72小时、1周的Caspase-3表达明显减少,差异具有统计学意义(P0.01),其它3组之间,差异无统计学意义(P0.05)。 结论 αB-晶状体蛋白对大鼠急性高眼压后视网膜神经节细胞存在保护作用,能增加视网膜神经节细胞的存活率。 αB-晶状体蛋白能抑制急性高眼压后视网膜神经节细胞内凋亡蛋白Caspase-3的表达,减少细胞凋亡。
[Abstract]:Objective to observe the effect of 伪 B crystallin on the survival of retinal ganglion cells and the expression of apoptotic protein Caspase-3 after acute intraocular hypertension in rats. Methods 3-month-old male Sprague-Dawley (SD) rats were used. 160 rats weighing 250 卤20 g were randomly divided into 4 groups: group A (acute intraocular pressure group) and group B (acute intraocular pressure vitreous puncture group) and group C (acute intraocular pressure vitreous injection normal saline group) and D group (acute high intraocular pressure group). The glass body was injected with 伪 B-crystallin group, and the fluorescent gold retrograde labeling method was used. The survival rate of retinal ganglion cells (RGCs) and the expression of apoptotic protein (Caspase-3) in retinal ganglion cells (RGCs) after acute intraocular pressure (IOP) were observed by immunofluorescence method at different time points. Results after acute high intraocular pressure, the retinal ganglion cells in the right eye (experimental eyes) of experimental animals began to decrease, and the total retinal ganglion cells decreased under low power fluorescence microscope. The decrease was most obvious in the first week and slowed down two weeks later. Compared with the other three groups, the survival rate of retinal ganglion cells in group D was significantly higher than that in group D at 1 week, 2 weeks and 3 weeks and 4 weeks after acute intraocular pressure (P0.01), and the survival rate of retinal ganglion cells in group D was significantly higher than that in group D (P0.01). There was no significant difference (P0.05) in the expression of Caspase-3 in the right eye (experimental eyes) of experimental animals after acute intraocular hypertension, and the expression of Caspase-3 in group D was significantly lower than that in group D (24 hours, 48 hours, 72 hours and 1 week after acute high intraocular pressure) compared with the other three groups. The difference was statistically significant (P0.01), the other three groups, the difference was not statistically significant (P0.05). Conclusion 伪 B-crystallin has protective effect on retinal ganglion cells after acute intraocular hypertension in rats. 伪 B-crystallin could inhibit the expression of apoptotic protein Caspase-3 in retinal ganglion cells after acute intraocular hypertension and decrease apoptosis.
