RECK和MMP-9在喉鳞癌中的表达及相关性研究

【摘要】:目的:探讨伴有kazal域的富含半胱氨酸的逆转诱导蛋白(revemion-inducing-cysteine- rich protein with kazal motifs, RECK)和基质金属蛋白酶-9(matrix metalloproteinase-9 MMP-9)在喉鳞状细胞癌和癌旁不同距离组织中的表达及其与临床的关系。 方法:采用逆转录聚合酶链反应(reverse transcriptase polymerase chain reaction,RT-PCR)法及免疫组化SP法检测38例喉鳞状细胞癌及其癌旁0.2cm、0.5cm、1.0cm组织中RECK和MMP-9的表达情况,并结合临床,分析其临床病理参数的关系。 结果:RECK在喉鳞癌组织、癌旁0.2cm、0.5cm及1.0cm的组织中表达强度及表达率依次增高,组间比较有显著统计学差异(P0.001);MMP-9在喉鳞癌组织、癌旁0.2cm、0.5cm及1.0cm的表达强度及表达率依次降低,组间比较有显著统计学差异(P0.001)。RECK和MMP-9在癌组织分别与癌旁0.5cm和1.0cm的组织比较及在癌旁0.2cm分别与癌旁0.5cm和1.0cm的组织比较,表达均有统计学差异(P0.05);但癌组织与癌旁0.2cm的组织中比较及癌旁0.5cm与癌旁1.0cm的组织中比较,表达均无统计学差异(P0.05)。RECK和MMP-9的表达呈负相关(P0.05)。喉鳞癌中不同临床分期、病理组织学分级,淋巴转移情况,二者的表达有统计学差异(P0.05)。 结论:RECK在喉鳞癌组织中呈阴性表达,而MMP-9在喉鳞癌组织中则为阳性表达;二者的表达呈负相关;RECK和MMP-9与喉鳞癌的临床分期、分化程度和淋巴转移情况有相关性;二指标的表达在癌旁0.5 cm处有明显的变化。
[Abstract]:Aim: to investigate the expression of cysteine-rich reverse inducible protein (revemion-inducing-cysteine- rich protein with kazal motifs, RECK) and matrix metalloproteinase-9 (matrix metalloproteinase-9 MMP-9) with kazal domain in laryngeal squamous cell carcinoma (LSCC) and adjacent tissues. Methods: reverse transcriptase polymerase chain reaction (reverse transcriptase polymerase chain reaction,RT-PCR) and immunohistochemistry (SP) were used to detect the expression of RECK and MMP-9 in 38 cases of laryngeal squamous cell carcinoma and its adjacent 0. 2 cm + 0. 5 cm ~ (-1). The relationship between the clinical pathological parameters and the clinical pathological parameters was analyzed. Results the expression intensity and expression rate of 1.0cm in laryngeal squamous cell carcinoma (LSCC) tissues were significantly higher than those in normal laryngeal squamous cell carcinoma tissues (P0.001). The expression intensity and expression rate of MMP-9 in laryngeal squamous cell carcinoma tissues were significantly lower than those in normal laryngeal squamous cell carcinoma tissues (P0.001). The expression intensity and expression rate of MMP 9 and 1.0cm in the adjacent tissues of laryngeal squamous cell carcinoma (LSCC) were lower than those in normal laryngeal squamous cell carcinoma (P0.001). There were significant differences between the two groups (P0.001) .Reck and MMP-9 were compared with 0.5cm and 1.0cm in cancer tissue and 0.5cm and 1.0cm in adjacent 0.2cm, respectively. But there was no significant difference in the expression of 0.5cm and 1.0cm between cancer tissues and adjacent 0.2cm tissues (P0.05). The expression of reck and MMP-9 were negatively correlated (P0.05). In laryngeal squamous cell carcinoma, different clinical stages, histopathological grading, lymphatic metastasis, the expression of statistical difference between them (P0.05). Conclusion the expression of fraction reck is negative in laryngeal squamous cell carcinoma and MMP-9 is positive in laryngeal squamous cell carcinoma, and the expression of reck and MMP-9 are negatively correlated with the clinical stage, differentiation and lymphatic metastasis of laryngeal squamous cell carcinoma. The expression of two markers was significantly changed at 0. 5 cm adjacent to cancer.
