苦参碱对人鼻咽癌CNE2细胞裸鼠移植瘤生长及凋亡相关基因表达的影响

【摘要】:目的:观察苦参碱对人鼻咽癌CNE2细胞裸鼠移植瘤生长及凋亡相关基因表达的影响。方法:20只裸鼠皮下接种CNE2,建立移植瘤模型,成瘤后,随机分成4组,每组5只。苦参碱高、低剂量组分别腹腔注射100、50mg/kg苦参碱,阳性对照组腹腔注射顺铂2 mg/kg,阴性对照组给予生理盐水。给药7次后,应用体内抑瘤实验测定抑瘤率,分别采用real-time PCR法和Western blot法检测移植瘤组织中bax、bcl-2、caspase-3 mRNA和蛋白的表达。结果:阳性对照组和苦参碱低、高剂量组抑瘤率分别为51.6%和26.4%、43.2%。4组间移植瘤组织中bax、bcl-2、caspase-3的mRNA和蛋白表达差异有统计学意义(F=3.751、16.113、3.918和26.500、9.771、12.255,P0.05);与阴性对照组比较,阳性对照组和苦参碱高、低剂量组bax mRNA和蛋白表达及阳性对照组、苦参碱高剂量组caspase-3 mRNA和蛋白表达上调(P0.05),bcl-2 mRNA和蛋白表达下调(P0.05)。结论:苦参碱可通过调节CNE2移植瘤组织中凋亡相关基因的表达抑制肿瘤的生长。
[Abstract]:Aim: to observe the effect of matrine on the growth and apoptosis-related gene expression of human nasopharyngeal carcinoma (NPC) CNE2 cells in nude mice. Methods: 20 nude mice were inoculated subcutaneously with CNE2, to establish transplanted tumor model. After tumorigenesis, 20 nude mice were randomly divided into 4 groups, 5 in each group. Matrine was injected intraperitoneally into 100g / kg matrine in low dose group, and normal saline was injected intraperitoneally in positive control group with cisplatin 2 mg/kg, negative control group. After seven times of administration, the tumor inhibition rate was measured by tumor inhibition test in vivo, and the expression of bax,bcl-2,caspase-3 mRNA and protein in transplanted tumor tissues were detected by real-time PCR and Western blot methods respectively. Results: the inhibitory rates of bax,bcl-2,caspase-3 in the positive control group and matrine group were 51.6% and 26.4%, respectively. There were significant differences in the expression of mRNA and protein in the transplanted tumor tissues between the two groups (F3. 751, 16.113, 3.918 and 26.500, 9.771, 12.255, P0.05), and compared with the negative control group, the positive control group and matrine group were higher than those in the negative control group, the positive control group and the matrine group were higher than those in the negative control group, the positive control group and the matrine group were significantly higher than those in the negative control group. The expression of bax mRNA and protein in low dose group and positive control group, the expression of caspase-3 mRNA and protein in high dose group of matrine was up-regulated (P0.05), and the expression of bcl-2 mRNA and protein was down-regulated (P0.05). Conclusion: matrine can inhibit the growth of CNE2 by regulating the expression of apoptosis-related genes.
