血小板反应蛋白-1在氧诱导视网膜病变小鼠视网膜中的表达及意义

【摘要】:目的:检测血小板反应蛋白-1(Thrombonspondin-1 TSP-1)在氧诱导视网膜病变模型小鼠(OIR)视网膜中的表达,探讨其在视网膜新生血管形成中的作用。 方法:随机选取7日龄C57BL/6J新生小鼠40只,分为模型组(20只)及正常对照组(20只)。模型组小鼠通过高氧诱导的方法建立氧诱导视网膜病变(OIR)的模型。于小鼠出生后第7、9、11日时两组各随机抽取5只小鼠,取视网膜组织采用RT-PCR法检测TSP-1mRNA的表达水平;并于小鼠出生后第11日时两组随机取5只小鼠,运用荧光造影视网膜铺片对视网膜新生血管进行形态学观察。 结果:(1)荧光造影视网膜铺片观察视网膜血管形态:出生后第11日时,正常组小鼠视网膜铺片显示视网膜血管分布呈均匀的网状结构,而模型组小鼠视盘周围可见大片无灌注区,视网膜大血管扩张,仅在周边见毛细血管分布,为典型的OIR病理Ⅰ期表现。 (2) RT-PCR检测TSP-1mRNA的结果:正常对照组小鼠视网膜组织中TSP-1mRNA表达水平各时间点之间表达差异无统计学意义(P0.05),模型组小鼠视网膜组织中TSP-1mRNA表达水平各时间点之间表达差异有统计学意义(P0.05)。出生后7日,模型组与对照组小鼠视网膜组织中TSP-1mRNA表达水平无明显差异(P0.05);出生后9日,模型组小鼠视网膜组织中TSP-1mRNA表达水平下降(P0.05);出生后11日模型组小鼠视网膜组织中TSP-1mRNA表达水平明显低于正常组(P0.05),且较9日时模型鼠下降。 结论:在新生小鼠OIR模型视网膜血管生长发育抑制期,视网膜组织中TSP-1mRNA的表达逐渐下降,提示TSP-1可能作为负调节因子在早期即参与视网膜新生血管的形成过程。
[Abstract]:Aim: to detect the expression of platelet reactive protein-1 (Thrombonspondin-1 TSP-1) in (OIR) retina of mice with oxygen-induced retinopathy and to explore its role in retinal neovascularization. Methods: forty 7 day old C57BL/6J newborn mice were randomly divided into two groups: the model group (n = 20) and the normal control group (n = 20). The (OIR) model of retinopathy induced by oxygen was established in the model group by hyperoxia induction. Five mice were randomly selected from each group on the 7th and 11th postnatal days, and the expression of TSP-1mRNA was detected by RT-PCR method in the retina of the two groups, and 5 mice in the two groups were randomly selected at the 11th day after birth. The retinal neovascularization was observed by fluorescence angiography. Results: (1) the retinal vascular morphology was observed by fluorescein angiography: at the 11th day after birth, the retina of normal mice showed a uniform reticular distribution of retinal blood vessels. In the model group, a large area of no perfusion was found around the optic disc, and the large retinal vessels were dilated, and the capillary distribution was observed only around the optic disc. (2) the results of TSP-1mRNA detection by RT-PCR: there was no significant difference in the expression of TSP-1mRNA between different time points in normal control group (P0.05). There was significant difference in the expression of TSP-1mRNA between different time points (P0.05). 7 days after birth, there was no significant difference in TSP-1mRNA expression between the model group and the control group (P0.05), but the expression level of TSP-1mRNA in the retinal tissue of the model group decreased 9 days after birth (P0.05). The expression of TSP-1mRNA in the retina of mice in the model group was significantly lower than that in the normal group (P0.05), and decreased after 9 days. Conclusion: the expression of TSP-1mRNA in retinal tissue decreased gradually during the period of retinal vascular growth inhibition in neonatal OIR model of mice, suggesting that TSP-1 may be involved in the formation of retinal neovascularization as a negative regulatory factor in the early stage of retinal neovascularization.
