WWOX、p53及Bcl-2在鼻咽癌中的表达及意义

【摘要】: 目的 检测WWOX、p53及Bcl-2 mRNA及其蛋白在鼻咽癌中的表达并分析其临床意义。方法 收集2009年6月至2009年10月间在我院耳鼻喉科初诊、有完整临床及病理资料的鼻咽癌患者鼻咽部组织61例,鼻咽粘膜慢性炎组织20例,用RT-PCR方法检测WWOX. p53及Bcl-2 mRNA表达情况,应用免疫组化方法检测相应组织的蛋白表达情况,并探讨WWOX、p53和Bcl-2的表达及意义。 结果 1. WWOX的表达情况 WWOX mRNA在鼻咽癌和鼻咽粘膜慢性炎组织中平均表达水平分别为0.69±0.34和0.89±0.49,WWOX蛋白在鼻咽癌和鼻咽粘膜慢性炎组织中阳性表达率分别为39.34%和70.00%,WWOX mRNA和蛋白在鼻咽癌组织中的表达低于鼻咽粘膜慢性炎组织,p0.05;WWOX蛋白在鼻咽癌Ⅲ-Ⅳ期的表达低于Ⅰ~Ⅱ期,p0.05;在有淋巴结转移的病例中WWOX蛋白的表达低于无淋巴结转移者,p0.05。分别对WWOX蛋白的阳性表达在鼻咽癌患者不同性别、不同年龄段和各病理分型中进行比较,差异无统计学意义,p0.05。 2.p53的表达情况 p53 mRNA在鼻咽癌和鼻咽粘膜慢性炎组织中平均表达水平分别是0.88±0.59和0.51±0.21,p53蛋白在鼻咽癌和鼻咽粘膜慢性炎组织中阳性表达率分别为80.33%和40.00%, p53 mRNA和蛋白在鼻咽癌组织中的表达高于鼻咽粘膜慢性炎组织,p0.05;p53蛋白在鼻咽癌Ⅲ-Ⅳ期的阳性表达明显高于鼻咽癌Ⅰ~Ⅱ期,p0.05;p53蛋白的阳性表达在有无淋巴结转移间的比较,差异无统计学意义,p0.05,分别对不同性别、不同年龄段和各病理分型中的阳性表达进行比较,差异无统计学意义,p0.05。 3.Bcl-2的表达情况 Bcl-2 mRNA在鼻咽癌和鼻咽粘膜慢性炎组织中表达水平分别是0.44±0.32和0.24±0.11,Bcl-2蛋白在鼻咽癌和鼻咽粘膜慢性炎组织中阳性表达率分别为85.25%和35.00%,Bcl-2 mRNA和蛋白在鼻咽癌组织中的表达高于鼻咽粘膜慢性炎组织,p0.05;Bcl-2蛋白的阳性表达在鼻咽癌ⅠⅡ期和Ⅲ~Ⅳ期间的比较,差异无统计学意义,分别对不同性别、不同年龄段、有无淋巴结转移及各病理分型中的阳性表达进行比较,差异无统计学意义,p0.05。 4. WWOX与p53及Bcl-2蛋白在鼻咽癌组织中表达的相关性 在鼻咽癌组织中,WWOX与p53表达呈负相关,差异具有统计学意义,p0.05。 在鼻咽癌组织中,WWOX与Bcl-2表达呈负相关,差异具有统计学意义,p0.05。 结论 1. WWOX mRNA及蛋白在鼻咽癌组织中呈低表达,与鼻咽癌的TNM分期、有无淋巴结转移有关,提示WWOX的异常表达可能参与鼻咽癌的发生、发展,且与其预后有关。 2. p53 mRNA及蛋白在鼻咽癌组织中呈高表达,与鼻咽癌的TNM分期有关,提示p53的异常表达可能参与鼻咽癌的发生、发展。 3. Bcl-2 mRNA及蛋白在鼻咽癌组织中呈高表达,与鼻咽癌的TNM分期、有无淋巴结转移、病理分型、性别和年龄无关,提示Bcl-2的异常是鼻咽癌变的较早期事件,与鼻咽癌的淋巴转移和预后关系可能不大。 4. WWOX与p53和Bcl-2在鼻咽癌组织中的表达呈负相关,提示WWOX与p53和Bcl-2在鼻咽癌的发生、发展中可能相互影响,共同或协同起作用。
