外周血NF-κB和TNF-α表达与甲状腺相关性眼病相关性研究

【摘要】：目的：甲状腺相关性眼病（Thyroid Associated Ophthalmopathy TAO）是一种与甲状腺疾病相关的器官特异性自身免疫性疾病，其发病机制尚不明确。目前研究已证明，多种细胞因子（Cytokines，CK）如干扰素（Interferon，IFN）、白细胞介素-1（Interleukin -1，IL-1）、IL-4、肿瘤坏死因子（Tumor necrosis factor，TNF）等参与TAO的发生与发展。核因子-κB(NCR Clear factor kappa B，NF-κB)是一具有多功能转录与调节作用的蛋白质因子，它能与多种细胞因子和粘附因子基因启动子部位发生结合，增强这些基因的转录和表达。本课题通过检测NF-κB以及TNF-α在TAO患者血液中的表达，分析其与TAO的相关性，为临床治疗提供实验基础。 方法：研究对象共160例，分为三组：TAO组54例、Graves病（Graves disease，GD）组52例和正常对照（CON）组54例。用酶联免疫吸附法(ELISA)检测血清中TNF-α的水平，蛋白印迹法（western blot）检测外周血中有核细胞中NF-κB的水平。 结果：①TAO组、GD组和CON组外周血血清中TNF-α浓度分别为6.157±0.653ng/ml、14.088±2.598ng/ml和2.398±0.326ng/ml；与正常对照组比较，TAO和GD组血清TNF-α浓度显著增高（P0.01；P0.01）；与GD组比较，TAO组血清TNF-α浓度显著降低（P0.01）。 ②TAO组、GD组和CON组外周血细胞核蛋白NF-κB的含量分别为0.334±0.490ng/ml、1.186±0.159ng/ml和0.077±0.0005ng/ml；与正常对照组比较，TAO和GD组外周血NF-κB表达水平增高（P0.01；P0.01）；与GD组比较，TAO组外周血NF-κB表达水平显著降低（P0.01）。 结论：①TAO和GD患者外周血中TNF-α和NF-κB的表达水平显著增高。②TAO患者外周血中TNF-α和NF-κB的高表达没有特异性。
[Abstract]:Objective: thyroid associated ophthalmopathy (Thyroid Associated Ophthalmopathy TAO) is an organ-specific autoimmune disease associated with thyroid disease. At present, many cytokines (Cytokines,CK), such as interferon (Interferon,IFN), interleukin-1 (Interleukin -1), IL-4, tumor necrosis factor (Tumor necrosis factor,TNF), have been proved to be involved in the pathogenesis and development of TAO. Nuclear factor- 魏 B (NCR Clear factor kappa (NF- 魏 B) is a multifunctional transcription and regulatory protein factor, which can bind to the promoter of many cytokines and adhesion factors, and enhance the transcription and expression of these genes. In this study, we detected the expression of NF- 魏 B and TNF- 伪 in the blood of TAO patients, analyzed their correlation with TAO, and provided the experimental basis for clinical treatment. Methods: one hundred and sixty patients were divided into three groups: TAO group (n = 54), Graves's disease (Graves disease,GD) group (n = 52) and normal control group (CON) group) (n = 54). Enzyme linked immunosorbent assay (ELISA) was used to detect the level of TNF- 伪 in serum and Western blot (western blot) was used to detect the level of NF- 魏 B in nucleated cells. Results: the serum TNF- 伪 concentrations in 1TAO group, GD group and CON group were 6.157 卤0.653 ng / ml, 14.088 卤2.598ng/ml and 2.398 卤0.326 ng / ml, respectively. Compared with the normal control group, the serum TNF- 伪 concentration in TAO and GD groups was significantly higher (P0.01, P0.01), and that in TAO group was significantly lower than that in GD group (P0.01). The contents of nuclear protein NF- 魏 B in 2TAO group, GD group and CON group were 0.334 卤0.490 ng / ml 1.186 卤0.159ng/ml and 0.077 卤0.0005 ng / ml, respectively. Compared with normal control group, the expression of NF- 魏 B in peripheral blood of TAO and GD groups was higher than that in GD group (P0.01), and the expression of NF- 魏 B in TAO group was significantly lower than that in GD group (P0.01). Conclusion: the expression of TNF- 伪 and NF- 魏 B in peripheral blood of patients with 1TAO and GD is significantly higher than that in patients with 2TAO, but the high expression of TNF- 伪 and NF- 魏 B in peripheral blood of 2TAO patients has no specificity.
