喉鳞状细胞癌中CyclinE和p27的表达和临床意义

【摘要】:目的研究CyclinE和p27在喉鳞状细胞癌(laryngeal squamous cell carcinoma,LSCC)中的表达及其相关性,并探讨其与LSCC患者病情进展之间的关系。 方法1、应用逆转录-多聚酶链反应(Reverse Transcription PCR, RT-PCR)技术对14例LSCC及癌旁正常喉黏膜组织(normal laryngeal tissues, NLT)中CyclinE和p27基因进行扩增,并检测上述组织中二者的mRNA水平;2、免疫组织化学Elivision染色法检测160例LSCC和对照组30例NLT中CyclinE和p27蛋白的表达水平,X2检验分析CyclinE和p27在LSCC及NLT的表达差异及其与LSCC临床病理因素之间的关系,Spearman等级相关分析CyclinE和p27之间的关系,P0.05为差异有统计学意义。 结果1、CyclinE和p27mRNA在LSCC和NLT中均有阳性表达。LSCC和NLT组织中CyclinE mRNA的相对表达量分别为1.8007±0.1063和0.8458±0.1230,组间比较差异有统计学意义(P0.05);p27mRNA的相对表达量分别为0.6510±0.1451和1.5216±0.1923,组间比较差异有统计学意义(P0.05);2、CyclinE和p27蛋白在LSCC和NLT中表达水平不同。LSCC和NLT组织中,CyclinE蛋白阳性率分别为64.37%(103/160)和16.67%(5/30); p27蛋白阳性率分别为40%(64/160)和60%(18/30),组间比较差异有统计学意义(P0.05),,LSCC中CyclinE和p27的表达与肿瘤分化程度、淋巴结转移和临床分期有关(P0.05),与年龄、性别及直径无关(P0.05);Spearman相关性分析显示CyclinE和p27之间呈显著负相关(R=-0.542,P0.05)。 结论: 1. CyclinE和p27mRNA在LSCC和NLT中均呈阳性表达,在LSCC中CyclinEmRNA显著上调,p27mRNA显著下降。 2. CyclinE蛋白在LSCC中高表达,在NLT中低表达甚至不表达;p27蛋白在LSCC中低表达甚至不表达,在NLT中呈高表达。 3. CyclinE和p27mRNA与CyclinE蛋白和p27蛋白在LSCC和NLT中表达一致。 4. CyclinE的表达水平与肿瘤分化程度、淋巴结转移和临床分期呈正相关,LSCC进展的重要指标。p27的表达水平与肿瘤分化程度、淋巴结转移和临床分期呈负相关,是LSCC进展的重要指标。
[Abstract]:Objective to investigate the expression of CyclinE and p27 in laryngeal squamous cell carcinoma (laryngeal squamous cell carcinoma,LSCC) and their correlation with the progression of LSCC. Methods 1. The CyclinE and p27 genes were amplified by reverse transcription-polymerase chain reaction (Reverse Transcription PCR, RT-PCR) in 14 cases of LSCC and adjacent normal laryngeal mucosa, and their mRNA levels were detected. 2. The expression of CyclinE and p27 protein in 160 cases of LSCC and 30 cases of control group were detected by immunohistochemical Elivision staining. The expression of CyclinE and p27 in LSCC and NLT and their relationship with the clinicopathological factors of LSCC were analyzed by X2 test. Spearman grade correlation analysis of the relationship between CyclinE and p27, P0.05 for the difference was statistically significant. Results 1CyclinE and p27mRNA were positive in LSCC and NLT, the relative expression of CyclinE mRNA in LSCC and NLT were 1.8007 卤0.1063 and 0.8458 卤0.1230respectively, the difference was statistically significant (P0.05). The relative expression of p27mRNA was 0.6510 卤0.1451 and 1.5216 卤0.1923, respectively, and the difference was statistically significant (P0.05). 2the expression levels of cyclin E and p27 protein in LSCC and NLT were different. The positive rates of CyclinE protein in LSCC and NLT were 64.37% (103 / 160) and 16.67% (5 / 30), respectively. The positive rates of p27 protein were 40% (64 / 160) and 60% (18 / 30), respectively. The difference between the two groups was statistically significant (P0.05). The expression of CyclinE and p27 in), LSCC was related to tumor differentiation, lymph node metastasis and clinical stage (P0.05). It was not related to age, sex and diameter (P0.05). Spearman correlation analysis showed that there was a significant negative correlation between CyclinE and p27 (P 0.05). Conclusion: 1. Both CyclinE and p27mRNA were positive in LSCC and NLT, CyclinEmRNA was up-regulated in LSCC, and p27mRNA was significantly decreased. 2. CyclinE protein was overexpressed in LSCC, low or non-expressed in NLT, and p27 protein was overexpressed in LSCC and NLT. 3. The expression of CyclinE and p27mRNA was consistent with that of CyclinE and p27 in LSCC and NLT. 4. The expression of CyclinE was positively correlated with the degree of tumor differentiation, lymph node metastasis and clinical stage. The expression level of p27 was negatively correlated with the degree of tumor differentiation, lymph node metastasis and clinical stage, and was an important indicator of the progression of LSCC.
