PTTG和VEGF-C在喉癌组织中的表达及其与微淋巴管生成的关系

【摘要】:目的探讨垂体肿瘤转化基因(PTTG)和血管内皮生长因子-C(VEGF-C)在喉癌组织中表达的相关性以及与喉癌发展的关系。方法应用免疫组织化学SP法分别检测60例喉癌组织和32例癌旁组织中PTTG和VEGF-C两种蛋白的表达情况,并用D2-40阳性产物定位淋巴管内皮细胞细胞质及细胞膜,计数微淋巴管密度(LMVD)。结果喉癌组织中PTTG和VEGF-C表达水平均显著高于癌旁组织,差异有统计学意义(P0.05);二者表达量与喉癌的分化程度、有无淋巴结转移及临床分期差异均有统计学意义(P0.05),与临床分型、有无吸烟史、肿瘤大小、性别及年龄方面的差异均无统计学意义(P0.05)。喉癌组织中LMVD表达水平明显高于癌旁组织,差异有统计学意义(P0.05),与淋巴结转移显著相关(P0.05),与分化程度、临床分期、临床分型、有无吸烟史、肿瘤大小、性别及年龄不相关(P0.05)。PTTG和VEGF-C在喉癌组织中表达呈显著正相关(P0.05)。LMVD分别与喉鳞癌组织中PTTG和VEGF-C蛋白表达正相关(P0.05)。结论 PTTG和VEGF-C在喉癌的发展中起重要作用,二者的检测可作为喉癌转移及预后的预判指标,有望成为新的基因治疗靶点。
[Abstract]:Aim to investigate the relationship between the expression of pituitary tumor transforming gene (PTTG) and vascular endothelial growth factor-C (VEGF-C) in laryngeal carcinoma and its relationship with the development of laryngeal carcinoma. Methods Immunohistochemical SP method was used to detect the expression of PTTG and VEGF-C proteins in 60 cases of laryngeal carcinoma and 32 cases of paracancerous tissues. Counting microlymphatic vessel density (LMVD). Results the expression levels of PTTG and VEGF-C in laryngeal carcinoma tissues were significantly higher than those in paracancerous tissues (P0.05). There was significant difference between the expression level and differentiation degree of laryngeal carcinoma, lymph node metastasis and clinical stage (P0.05), as well as clinical classification, smoking history, tumor size, and so on. There was no significant difference in sex and age (P0.05). The expression level of LMVD in laryngeal carcinoma was significantly higher than that in paracancerous tissues (P0.05), and correlated with lymph node metastasis (P0.05), differentiation degree, clinical stage, clinical classification, smoking history, tumor size, and so on. There was a significant positive correlation between the expression of PTTG and VEGF-C in laryngeal carcinoma (P0.05). LMVD was positively correlated with the expression of PTTG and VEGF-C protein in laryngeal squamous cell carcinoma (P0.05). Conclusion PTTG and VEGF-C play an important role in the development of laryngeal carcinoma. The detection of them can be used as predictors of metastasis and prognosis of laryngeal carcinoma and may be a new target of gene therapy.
