变应性鼻炎中Th17细胞功能测定及临床意义

【摘要】: 目的 观察一种新型的CD4+效应T细胞——辅助性17细胞(T help 17, Th17)及其相关免疫学细胞因子在变应性鼻炎中的表达情况,以探讨其在变应性鼻炎发病机理中的作用,并为进一步研究Th17细胞的功能调控机制奠定基础。 方法 选择30例变应性鼻炎伴下鼻甲肥大或伴鼻息肉患者作为病例组,25例鼻中隔偏曲并下鼻甲肥大患者作为对照组,于鼻内镜手术过程中严格无菌操作取鼻粘膜。采用免疫组织化学法进行染色检测病例组和对照组Th17的表达水平;通过ELISA法检测IL-17、TGF-β在病例组和对照组的表达情况,最后进行统计学分析。 结果 1.Th17阳性细胞的平均水平变应性鼻炎患者组和对照组存在明显差异(P0.001),Th17在变应性鼻炎患者鼻粘膜高表达,对照组鼻粘膜低表达。 2.IL-17及TGF-p在变应性鼻炎患者组和对照组存在明显差异(P0.001),变应性鼻炎患者组比对照组高。 3.变应性鼻炎患者组Th17与TGF-p存在正相关(P0.001)。 结论 1.变应性鼻炎患者中Th17细胞的高表达可能是一个重要原因。 2.TGF-β可能协同Th17细胞参与变应性鼻炎的发生发展。 3.在变应性鼻炎中,Th17细胞TGF-β之间密切相关,或许可以为变应性鼻炎的治疗提供一条新思路。
[Abstract]:Objective to observe the expression of helper 17 cell (T help 17 (Th17) and its related immunological cytokines in allergic rhinitis (AR), a new type of CD4 effector T cell. In order to explore its role in the pathogenesis of allergic rhinitis, and lay a foundation for further study of the functional regulation mechanism of Th17 cells. Methods 30 patients with allergic rhinitis with inferior turbinate hypertrophy or nasal polyps were selected as the case group and 25 patients with nasal septum deviation and inferior turbinate hypertrophy as the control group. The nasal mucosa was obtained strictly during endoscopic surgery. Immunohistochemical staining was used to detect the expression of Th17 in the case group and control group, and ELISA method was used to detect the expression of IL-17,TGF- 尾 in the case group and the control group. Results the average level of 1.Th17 positive cells was significantly different between the allergic rhinitis group and the control group (P0.001). The expression of Th17 was high in the nasal mucosa of the allergic rhinitis patients, and low in the nasal mucosa of the control group. There was a significant difference in 2.IL-17 and TGF-p between the allergic rhinitis group and the control group (P0.001), and it was higher in the allergic rhinitis group than that in the control group. 3. There was a positive correlation between Th17 and TGF-p in patients with allergic rhinitis (P0.001). Conclusion 1. The high expression of Th17 cells in patients with allergic rhinitis may be an important cause. 2. TGF-尾 may cooperate with Th17 cells in the development of allergic rhinitis. 3. In allergic rhinitis, TGF- 尾 of Th17 cells is closely related, which may provide a new idea for the treatment of allergic rhinitis.
