NT-3对哇巴因损伤耳蜗螺旋神经节细胞后的影响

【摘要】:目的: 建立畦巴因选择性损伤蒙古沙鼠耳蜗螺旋神经节细胞的动物模型,在此基础上研究耳蜗螺旋神经节细胞损伤后,神经营养因子-3对耳蜗螺旋神经节细胞的保护作用,并研究探讨其神经保护作用机理。 方法: 将听力正常的雄性蒙古沙鼠,手术暴露圆窗膜,用80umol/L哇巴因滴注于蒙古沙鼠耳蜗圆窗膜上,建立选择性损伤螺旋神经节细胞的动物模型。在成功建模的基础上于蒙古沙鼠圆窗膜上滴注神经营养因子-3(15ug/ml),稳定7天后处死,立即取出耳蜗,制作病理切片进行HE染色,观察Ⅰ型螺旋神经节细胞数目、形态的改变,并用人工计数法采用SPSS17.0软件进行统计学分析;制作电镜切片,在透射电镜下观察细胞的超微结构,分析神经营养因子-3对Ⅰ型螺旋神经节细胞的保护作用。 结果: 本实验用哇巴因成功建立了选择性损伤螺旋神经节细胞的动物模型,发现哇巴因对Ⅰ型螺旋神经节细胞有明显的损伤作用,损伤部位主要位于细胞膜周围髓鞘组织、细胞核及线粒体,哇巴因组细胞计数较人工外淋巴液组明显减少(P0.05);神经营养因子-3对Ⅰ型螺旋神经节细胞的细胞膜周围髓鞘组织、线粒体及细胞核均有保护作用,在保护组细胞计数明显高于对照组(P0.05)。 结论: 哇巴因损伤蒙古沙鼠Ⅰ型螺旋神经节细胞后,经圆窗膜滴注神经营养因子-3(NT-3)对Ⅰ型螺旋神经节细胞有一定的保护作用。对于临床上寻找保护耳蜗听神经细胞的有效药物及给药途径具有重要的价值。
