通过大鼠凝血功能实验探讨消栓通脉饮对华法林钠抗凝作用的影响

【摘要】：目的:探讨中药消栓通脉饮对华法林钠抗凝作用的影响,指导临床中西医结合治疗下肢深静脉血栓形成安全用药。方法:建立下肢深静脉血栓形成大鼠模型,分为消栓通脉饮组、华法林钠组、消栓通脉饮与华法林钠合用组、模型组和空白对照组,于造模后第二天开始分别给予消栓通脉饮、华法林钠、消栓通脉饮与华法林钠、生理盐水灌胃,连续给药7天,分别于大鼠用药后96h及192h腹主动脉取血、抗凝,测定大鼠用药后96h及192h活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(FIB)、凝血酶原时间(PT)和国际标准化比值(INR)等指标。结果:给药96h和192h后INR值、PT值、APTT值、FIB值和TT值,模型组较空白组有显著的变化(P0.05),华法林钠组用药时间不同各项指标不稳定,消栓通脉饮组及联合用药组各项指标较稳定。结论:华法林钠与消栓通脉饮均有抗凝作用。华法林钠抗凝作用显著,但有较明显的不良反应,较易引起出血倾向;消栓通脉饮抗凝作用稳定,不易引起出血倾向;消栓通脉饮能够稳定华法林钠的抗凝作用,控制华法林钠引起的出血倾向。
[Abstract]:Objective: to explore the effect of Xiaoshuantongmai decoction on warfarin sodium anticoagulant effect, and to guide the safe treatment of lower extremity deep vein thrombosis with integrated Chinese and western medicine. Methods: to establish a rat model of deep vein thrombosis of lower extremity. They were divided into two groups: Xiaoshuantongmai decoction group, warfarin sodium group, Xiaoshuantongmai decoction combined with warfarin sodium group, model group and blank control group. Xiaoshuantongmai Yin, warfarin sodium and normal saline were perfused by stomach for 7 days. Blood was taken from abdominal aorta of rats 96 h and 1922 h, respectively. The activated partial thromboplastin time (APTTT), thrombin time (TTT) and fibrinogen (FIBs) were measured at 96 h and 1922 h after administration. Results: after 96 h and 1922 h of administration, the INR value, PT value, APTT value, FIB value and TT value were determined. Compared with the blank group, the model group had a significant change (P 0.05), and the warfarin sodium group was unstable in different time. Conclusion: warfarin sodium and Xiaoshuantongmai decoction have anticoagulant effect, warfarin sodium has obvious anticoagulant effect, but there are obvious adverse reactions. It is easy to cause bleeding tendency; The anticoagulant effect of Xiaoshuan Tongmai Yin is stable and it is not easy to cause bleeding tendency. Xiaoshuan Tongmai decoction can stabilize the anticoagulant effect of warfarin sodium and control the bleeding tendency caused by warfarin sodium.
