林肯链霉菌高产菌株选育研究

【摘要】:为进一步提高林可霉素生产发酵水平,本文以林肯链霉菌LK15-35为出发菌株,采用氯化锂(Li Cl)、常压室温等离子体(ARTP)和紫外线(UV)三重复合诱变,首先用Li Cl预处理4h,然后用ARTP照射60s,再用UV照射30s,最终获得高产突变株LK16-77。结果表明,摇瓶发酵单位提高30.0%,经180吨罐生产验证,平均放罐效价提高21.4%。稳定性传代验证证明该突变株高产性能遗传稳定。
[Abstract]:In order to further improve the fermentation level of lincomycin, Streptomyces Lincoln LK15-35 was used as the starting strain. The high yield mutant LK16-77 was obtained by pretreatment with Li Cl for 4 h, then irradiated with ARTP for 60 s and then irradiated with UV for 30 s. The results showed that the fermentation unit of shaking flask was increased by 30.0% and verified by 180 ton pot production. The average titer of the mutant was increased by 21.4.The stability test proved that the mutant was genetically stable in high yield.
