MK-801急性处理改变斑马鱼幼鱼自发运动和焦虑行为

【摘要】:目的在不同光照条件下,地卓西平马来酸盐(dizocilpine,MK-801)急性处理对斑马鱼幼鱼自发运动和焦虑行为的影响。方法野生型AB品系斑马鱼幼鱼(5天和7天)暴露在不同浓度MK-801(0、10、20、50、100、200μmol/L)溶液中。在光照与黑暗交替条件下,记录斑马鱼幼鱼每分钟游动距离和趋触性。结果光照条件下,MK-801急性处理能显著降低幼鱼自发运动(P0.05);黑暗条件下,MK-801急性处理也能降低幼鱼自发运动(P0.05),但10μmol/L MK-801对5天幼鱼自发运动无显著影响(P0.05);黑暗条件下,7天幼鱼在100μmol/L MK-801作用下抗焦虑能力显著增加,5天幼鱼在20、50μmol/L MK-801作用下有显著的抗焦虑能力(P0.05);光照条件下,斑马鱼幼鱼的抗焦虑行为都无显著变化(P0.05)。结论 MK-801诱导幼鱼自发运动降低;在黑暗条件下,MK-801用药组(20、50、100和200μmol/L)的幼鱼表现出抗焦虑行为;年龄、药物剂量和光照条件共同作用影响斑马鱼幼鱼的行为模式。
[Abstract]:Objective under different illumination conditions, The effect of dizocilpine maleate on spontaneous movement and anxiety behavior of juvenile zebrafish was studied. Methods Wild AB strain of juvenile zebrafish was exposed to MK-801 (102050U / L) solution for 5 days and 7 days. Under the alternating conditions of light and darkness, the effect of dizocilpine on the spontaneous movement and anxiety behavior of juvenile zebrafish was studied. The swimming distance and thixotropism per minute of juvenile zebrafish were recorded. Results under light condition, acute treatment with MK-801 significantly reduced the spontaneous movement of young fish P0.05U, and acute treatment with MK-801 also decreased the spontaneous movement of juvenile fish P0.05, but 10 渭 mol/L MK-801 for 5 days. There was no significant effect on the spontaneous movement of young fish (P0.05), but the ability of anti-anxiety was significantly increased under 100 渭 mol/L MK-801 for 7 days under dark conditions, and the ability of anti-anxiety was significantly increased under the action of 2050 渭 mol/L MK-801 at 20 渭 mol/L MK-801 for 5 days. There was no significant change in the anti-anxiety behavior of juvenile zebrafish (P0.050.Conclusion MK-801 induces the decrease of spontaneous movement of juvenile fish, and the juvenile of MK-801 drug group shows anti-anxiety behavior under dark conditions, which is 200 渭 mol / L and 20 渭 mol / L). The effects of drug dosage and light condition on the behavior patterns of juvenile zebrafish were studied.
