塞来昔布体外药物作用机制的相关研究

【摘要】:目的观察塞来昔布体外实验效应及相关影响因素,探索其体外药物作用的相关机制。方法利用MTT比色法测定塞来昔布对多种细胞系、药物作用不同时间以及与细胞短时间接触后产生的抑制率;通过测定血红蛋白的吸光度来反应不同条件下药物引起的溶血作用;观察不同培养基血清浓度、药物配置后放置不同时间、超声及滤膜过滤等物理处理方式对药物体外作用的影响。结果体外培养基条件下,塞来昔布可同时抑制肿瘤细胞系及正常细胞系的生长,药物的这种作用在与药物短暂接触后即会产生,且药物抑制作用会随作用时间的延长而逐渐增强;较高浓度药物可在短时间内引起红细胞胞膜破裂,这种作用可在4℃的环境下中止,恢复温度后红细胞再次破裂;培养基中血清浓度、药物配置后放置时间以及超声和滤膜过滤等因素均可明显影响药物的体外抑制作用。结论培养基中未溶解的游离形态药物是塞来昔布体外效应的作用主体,其通过快速黏附于细胞膜系统而产生一系列特殊的实验现象,故可影响药物游离形式的方式均可明显影响药物的体外效应。
[Abstract]:Objective to observe the experimental effect of celecoxib in vitro and its related influencing factors, and to explore the mechanism of its drug action in vitro. Methods the effects of celecoxib on various cell lines were determined by MTT colorimetry. Inhibition rate of drugs at different time and short contact with cells; hemolysis induced by drugs under different conditions by measuring the absorbance of hemoglobin; observation of serum concentration in different media, The effects of ultrasound and filter membrane filtration on the drug action in vitro were studied. Results Celecoxib could inhibit the growth of tumor cell line and normal cell line at the same time. This effect of the drug will be produced after brief contact with the drug, and the inhibitory effect of the drug will gradually increase with the prolongation of the action time, and the higher concentration of the drug can cause the rupture of the erythrocyte membrane in a short time.
