过渡金属铜修饰的仲钨酸盐抑制人卵巢癌细胞增殖的研究

【摘要】:研究了过渡金属铜修饰的仲钨酸盐化合物[Na_2(H_2O)_(10)][Cu_4(H_2O)_(12)(H_2W_(12)O_(42))]·15H_2O(CuW_(12))的体外抗肿瘤活性.取人卵巢癌细胞SKOV-3加入不同浓度CuW_(12)处理,应用四甲基偶氮唑盐(MTT)比色法分析细胞增殖,计算了半数抑制浓度(IC_(50));采用光学显微镜观察SKOV-3细胞的凋亡形态变化;分析了细胞周期和细胞凋亡,计算了各期细胞比例及细胞凋亡率.结果表明:CuW_(12)对SKOV-3细胞增殖抑制的IC_(50)值为45.2μmol/L,且呈剂量依赖性;与经典的仲钨酸盐Na_(10)(H_2W_(12)O_(42))·26H_2O(NaW_(12))相比,过渡金属铜修饰的仲钨酸盐抗肿瘤活性更强;光学显微镜下观察到CuW_(12)处理组细胞数目明显减少,出现变圆、缩小、老化等形态变化;不同浓度CuW_(12)处理12h后早期凋亡细胞所占的百分比显著增加且呈剂量依赖性.
[Abstract]:The in vitro antitumor activity of the transition-metal copper modified paratungstate compound [Na _ 2H _ S _ T _ 2O _ 10] [Cu4H _ S _ 2O _ O _ T] [Cu4H _ S _ 2H _ 2O _ O _ T _ T _ (12 / S))] 15H _ 2O-Stam _ CuW _ (12) was studied, and the cell proliferation was analyzed by MTT colorimetric analysis by adding different concentrations of CuW _ (12) to the human ovarian cancer cell line SKOV-3. The half inhibitory concentration (ICS) was calculated, the morphological changes of SKOV-3 cells were observed by optical microscope, and the cell cycle and apoptosis were analyzed. The cell proportion and apoptosis rate of each phase were calculated. The results showed that the ICSP 50) value of the inhibition of SKOV-3 cell proliferation was 45.2 渭 mol / L in a dose-dependent manner, and the antitumor activity of commatungstate modified by transition metal copper was higher than that of the classical secatungstate Nastra (42D) 26H _ 2O NaW _ (12) (P < 0.05). It was observed under optical microscope that the number of cells in the cuw _ (12) treatment group was obviously decreased, and the morphological changes such as circle, reduction and aging were observed, and the percentage of early apoptotic cells increased significantly after 12 h treatment with different concentrations of CuW _ (12) in a dose-dependent manner.
