天冬氨酸对脂多糖诱导的仔猪肝脏损伤和肌肉蛋白质降解的调控作用

【摘要】:本文研究了天冬氨酸(ASP)对脂多糖(LPS)刺激仔猪肝脏损伤和肌肉蛋白质降解的调控作用及其机制。 1、本试验研究了ASP对LPS刺激仔猪肝脏损伤的保护作用,并从Toll样受体4(TLR4)和核苷酸结合寡聚域受体(NOD)信号通路的角度探讨其机制。选用48头断奶仔猪,分为4个处理组:(1)对照组(基础日粮);(2)LPS组(基础日粮+LPS);(3)0.5%ASP组(基础日粮+0.5%ASP+LPS);(4)1.0%ASP组(基础日粮+1.0%ASP+LPS)。试验期为21天。第20天,给2、3和4组注射100μg/kg体重的LPS,1组注射等量的生理盐水,,4h和24h后采血。采血后屠宰,取肝脏样品待测。结果表明:(1)0.5%和1.0%ASP可缓解注射LPS4h和24h后肝脏形态结构的损伤。(2)1.0%ASP可缓解注射LPS4h后血清碱性磷酸酶(AKP)和γ-谷氨酸转肽酶活性升高;0.5%或1.0%ASP可缓解注射LPS24h后血清谷草转氨酶、AKP活性的升高和谷丙转氨酶/谷草转氨酶比例的降低(P0.05)。(3)1.0%ASP可缓解注射LPS24h后claudin-1蛋白表达量的降低(P0.05)。(4)0.5%和1.0%ASP缓解了注射LPS4h后肿瘤坏死因子-α(TNF-α),环氧合酶2(COX2)和热休克蛋白70mRNA表达量的升高,1.0%ASP缓解了注射LPS24h后TNF-α和COX2mRNA表达量的降低(P0.05)。(5)0.5%或1.0%ASP缓解了注射LPS4h后TLR4和其下游信号分子髓样分化因子88、白介素受体相关激酶1和肿瘤坏死因子受体相关因子6,以及NOD1、NOD2和其下游信号分子受体互作蛋白激酶2mRNA表达量的上升,降低了核因子(NF)-κBp65的mRNA表达量(P0.05);0.5%或1.0%ASP缓解了注射LPS24h后TLR4,NOD1和NOD2的mRNA表达量的降低(P0.05)。这些结果表明:在LPS刺激早期和晚期,ASP对TLR4和NOD信号通路相关基因的mRNA表达、炎性介质的产生发挥相反的调控作用,并改善了肝脏结构和功能。 2、本试验研究了ASP对LPS刺激仔猪肌肉蛋白质降解的保护作用,并从AMP激活蛋白激酶(AMPK)、蛋白激酶B/叉头转录因子(Akt/FOXO)、TLR4和NOD信号通路的角度探讨其机制。选用24头断奶仔猪,分为4个处理组:(1)对照组(基础日粮);(2)LPS组(基础日粮+LPS);(3)0.5%ASP组(基础日粮+0.5%ASP+LPS);(4)1.0%ASP组(基础日粮+1.0%ASP+LPS)。试验期为19天。第19天,给2、3和4注射100μg/kg体重的LPS,1组注射等量的生理盐水,4h后屠宰,取肌肉样品待测。结果表明:(1)0.5%ASP提高了腓肠肌DNA和RNA含量(P0.05);1.0%ASP提高了腓肠肌蛋白质和RNA含量,提高了背最长肌RNA/DNA比值和蛋白质/DNA比值(P0.05)。(2)1.0%ASP缓解了注射LPS后腓肠肌肌萎缩F-box(MAFbx)和肌肉环指蛋白1(MuRF1)mRNA的表达量上升(P0.05),0.5%和1.0%ASP缓解了注射LPS后背最长肌MuRF1mRNA的表达量上升(P0.05)。(3)0.5%或1.0%ASP降低了腓肠肌pAMPKα/tAMPKα,缓解了注射LPS后肌肉pAkt/tAkt和pFOXO1/tFOXO1降低,降低了背最长肌FOXO4的mRNA表达量(P0.05)。(4)ASP没有抑制肌肉TLR4信号通路相关基因的表达,相反还提高了背最长肌簇分化抗原14、MD2和脂多糖结合蛋白的mRNA表达量(P0.05)。(6)1.0%ASP提高了腓肠肌的细胞因子信号抑制物1(SOCS1)和toll反应蛋白(Tollip),背最长肌的RP105、SOCS1、Tollip和单免疫球蛋白IL-1受体相关分子的mRNA表达量(P0.05)。这些结果表明:ASP不是通过TLR4和NOD信号通路,而是通过抑制AMPKα磷酸化,提高Akt磷酸化,进而提高FOXO磷酸化,从而抑制MAFbx和MuRF1表达来缓解肌肉蛋白质降解。
[Abstract]:In this paper , the regulation and mechanism of lipopolysaccharide ( ASP ) on liver injury and muscle protein degradation in piglets stimulated by lipopolysaccharide ( LPS ) were studied .

1 . The protective effect of ASP on LPS - stimulated liver injury in piglets was studied , and its mechanism was discussed from the angle of Toll - like receptor 4 and nucleotide binding oligopeptide signal pathway . 48 weaned piglets were divided into 4 treatment groups : ( 1 ) control group ( basal diet ) ;
( 2 ) LPS group ( basal diet + LPS ) ;
( 3 ) 0.5 % ASP group ( basal diet + 0.5 % ASP + LPS ) ;
( 4 ) 1.0 % ASP group ( basal diet + 1.0 % ASP + LPS ) .
( 3 ) The levels of TNF - 伪 , COX2 and HSP70 mRNA were decreased after LPS24 injection ( P0.05 ) . ( 5 ) 0.5 % or 1.0 % ASP alleviated the increase of TNF - 伪 and COX2 mRNA expression after injection of LPS24 .
The results showed that after LPS stimulated early and late LPS stimulation , ASP had the opposite effect on the mRNA expression and inflammatory mediators , and improved the structure and function of liver .

2 . This study investigated the protective effects of ASP on muscle protein degradation in piglets stimulated by LPS , and discussed its mechanism from the angle of AMP - activated protein kinase ( AMPK ) , protein kinase B / protein kinase B / protein kinase B / protein kinase B / protein kinase B / protein kinase B / FOXO . The 24 - head weaned piglets were divided into 4 treatment groups : ( 1 ) control group ( basal diet ) ;
( 2 ) LPS group ( basal diet + LPS ) ;
( 3 ) 0.5 % ASP group ( basal diet + 0.5 % ASP + LPS ) ;
( 4 ) 1.0 % ASP group ( basal diet + 1.0 % ASP + LPS ) . The test period was 19 days . On day 19 , 100 渭g / kg body weight of LPS was injected into 2 , 3 and 4 groups of LPS , 1 group was injected with equal amount of physiological saline . After 4 hours , the muscle samples were slaughtered . The results showed that : ( 1 ) 0.5 % ASP increased the DNA and RNA content of calf muscle ( P0.05 ) ;
( 2 ) 1 . 0 % ASP reduced the mRNA expression level of the muscle atrophy F - box and the muscle ring finger protein 1 ( P0.05 ) .
