右美托咪定预处理对缺血再灌注大鼠心肌细胞的保护作用

【摘要】:目的研究右美托咪定(Dex)预处理对缺血再灌注大鼠心肌细胞凋亡及凋亡蛋白表达的影响。方法按照体重将雄性SD大鼠随机分为4组,每组7只:低高2个剂量实验组(结扎前0.5 h分别腹腔注射Dex 10,30μg·kg~(-1)),模型组及假手术组(结扎前0.5 h均腹腔注射0.9%Na Cl 5 m L·kg~(-1))。用结扎左冠状动脉前支构建心肌缺血-再灌注损伤大鼠模型。以缺口末端标记(TUNEL)法检测各组大鼠心肌细胞凋亡率;以免疫组化法检测各组大鼠心肌组织中Bcl-2及Bax蛋白表达情况。结果假手术组、模型组与低高2个剂量实验组的心肌细胞凋亡率(AR,%)分别为0.42±0.06,19.73±4.12,13.31±3.65,10.04±2.71;这4组的心肌组织Bcl-2阳性表达指数(PEI)分别为0.25±0.03,5.74±1.06,11.44±2.01,14.71±2.13;这4组的心肌组织Bax PEI分别为0.27±0.04,19.95±5.01,15.02±4.82,12.13±3.14;这4组的Bcl-2/Bax分别为0.93±0.05,0.29±0.04,0.76±0.06,1.20±0.07。与假手术组比较,模型组大鼠心肌细胞AR、Bcl-2、Bax PEI及Bcl-2/Bax的差异均有统计学意义(均P0.01);与模型组比较,低高2个剂量实验组大鼠心肌组织这4项指标差异均有统计学意义(均P0.01)。结论 Dex可通过上调Bcl-2蛋白表达及下调Bax蛋白表达,抑制心肌缺血-再灌注损伤大鼠心肌细胞的凋亡。
[Abstract]:Aim to study the effects of dexmetomidine Dexpreconditioning on myocardial apoptosis and apoptotic protein expression in rats with ischemia reperfusion.Methods male Sprague-Dawley rats were randomly divided into 4 groups according to their body weight.
