重组人卵泡刺激素类似物对幼龄小鼠卵巢发育的影响

【摘要】:目的:探讨重组人卵泡刺激素(recombinant follicle stimulating hormone,r FSH)类似物对幼龄小鼠卵巢发育的影响。方法:3周龄小鼠随机分为6组:溶媒空白对照组、果纳芬单次注射组(0.5 IU)和果纳芬3次注射组(0.5 IU·d-1,连续给药3 d);r FSH类似物低剂量组(0.375 IU)、中剂量组(0.75 IU)和高剂量组(1.5 IU)均在停药24 h后摘取卵巢称重并计算卵巢指数,卵巢组织经HE染色,观察各级卵泡所占比例。结果:与空白对照组相比,r FSH类似物能剂量依赖性地促进卵巢增重,显著增加卵巢指数(P0.05)。0.375 IU剂量即可显著促进幼龄小鼠卵巢指数的增加(P0.05),1.5 IU剂量组较阳性药物果纳芬3次注射组无显著性差异;0.75 IU r FSH类似物较空白对照组可显著降低原始卵泡所占比例,提高成熟卵泡和黄体所占比例(P0.01),且1.5 IU剂量组较阳性药物果纳芬3次注射组能更有效地降低原始卵泡的比例,提高成熟卵泡和黄体所占比例(P0.01)。结论:r FSH类似物不仅能够促进幼龄小鼠卵巢增重以及促进原始卵泡、初级卵泡和次级卵泡的发育,而且具长效性,即单次注射即可使血药浓度维持3 d。
[Abstract]:Aim: to investigate the effects of recombinant follicle stimulating hormoney analogue on ovarian development in young mice. Methods Twenty three week-old mice were randomly divided into 6 groups: solvent blank control group. In the single injection group (0.5 IUU) and the 3 times injection group (0.5 IU d-1), the low dose group (0.375 IUU) and the middle dose group (0.75 IUU) and the high-dose group (1.5-IUU) were taken out 24 hours after the withdrawal of the drug and the ovarian index was calculated. The proportion of follicles was observed by HE staining in ovarian tissues. Results: compared with the control group, the analogue of r FSH could promote ovarian weight gain in a dose-dependent manner. The ovarian index of young mice was significantly increased by increasing the dosage of P0.05U. 0.375IU. The dose of P0.05U 1.5IU was significantly lower than that of the three injection group of positive drug, the analogue of 0.75IU r FSH was significantly lower than that of the blank control group. Proportion of low primordial follicles, The proportion of mature follicles and corpus luteum was increased, and the proportion of mature follicles and corpus luteum was increased more effectively in 1.5 IU group than that in the three injection groups of positive drug Funafen, and the proportion of mature follicles and corpus luteum was increased. Conclusion the fraction r FSH analogue can not only promote ovarian weight gain and the development of primordial follicles, primary follicles and secondary follicles in young mice, but also have a long-term effect, that is, a single injection can maintain the blood concentration for 3 days.
