基于多胺转运系统的抗肿瘤药物研究进展

【摘要】：多胺是维持细胞生长分化等生命活动的必需物质。肿瘤的增殖与细胞内多胺水平密切相关,利用肿瘤细胞膜表面高表达的多胺转运系统介导多胺类似物和缀合物进入细胞,可耗竭细胞内多胺并诱导肿瘤细胞凋亡。也可利用多胺聚阳离子的结构特点进行基因递送,提高外源基因的转染效率。综述了基于多胺转运系统的多胺类似物,缀合物和基因递送的癌症治疗研究进展,旨为以多胺结构为基础的抗肿瘤药物研究提供参考。
[Abstract]:Polyamines are essential substances to maintain cell growth and differentiation. The proliferation of tumor is closely related to the level of intracellular polyamines. Polyamine analogues and conjugates are mediated into the cells by the highly expressed polyamine transport system on the surface of the tumor cell membrane, which can deplete the polyamines in the cells and induce apoptosis of the tumor cells. The structure of polyamine polycation can also be used for gene delivery to improve the efficiency of foreign gene transfection. The progress in cancer treatment of polyamine analogues, conjugates and gene delivery based on polyamine transport system is reviewed in this paper, which provides a reference for the study of antitumor drugs based on polyamine structure.
