环境中低浓度四溴双酚A对颤蚓的毒性研究

【摘要】:本文以颤蚓为受试生物,研究四溴双酚A(TBBPA)对颤蚓的毒性效应以及颤蚓对TBBPA的富集效果。首先通过急性毒性实验测得TBBPA对颤蚓的96 h—LC50;其次根据急性毒性试验结果进行半静态试验,选取0、0.025、0.05、0.25、0.50和1.0 mg·L-1六组浓度进行染毒实验,再选取1 mg·L-1浓度培养1、3、5、7和10 d后,测定各组颤蚓体内酶活性;最后设置TBBPA高浓度(1 mg·L-1)和低浓度(0.2 mg·L-1)两组染毒7 d后,测定颤蚓对TBBPA的富集系数(BCF)。主要结果如下:1.随着暴露浓度和暴露时间的增加,各处理组颤蚓的死亡率上升,TBBPA毒性明显增强,存在明显剂量—效应关系。TBBPA对颤蚓48 h、72 h、96 h的LC50分别为11.295 mg·L-1、3.864 mg·L-1、2.204 mg·L-1,安全浓度为0.22 mg·L-1。2.暴露1 d和7 d后,颤蚓体内的超氧化物岐化酶(SOD)表现为诱导效应,其中0.05 mg·L-1浓度组诱导最大(P0.01),分别为对照组的1.36和1.61倍;过氧化氢酶(CAT)活性以诱导效应为主,同样在0.05 mg·L-1浓度组达到峰值(P0.01),但暴露7 d的CAT活性总体低于暴露1 d的CAT活性;过氧化物酶(POD)活性呈现先诱导后抑制的变化趋势,在0.025 mg·L-1浓度组即达到最大诱导(P0.01),分别为对照组的1.28和1.38倍,在1 mg·L-1浓度组时活性降至最低,抑制率分别为8%和20%;乙酰胆碱酯酶(AChE)活性先增加后降低,各处理组AChE活性均显著高于对照组(P0.01),当TBBPA浓度为0.25 mg·L-1时达到峰值,分别为对照组的1.25和1.20倍。3.在1 mg·L-1TBBPA暴露的10 d内,SOD、CAT和AChE活性均出现先升高后下降的变化趋势,其活性分别在第3 d和第5 d达到最大值(P0.01)。POD的变化则分为降低、升高、再降低3个阶段,其活性在第3 d到达峰值(P0.01)。4.随着染毒液浓度的升高,水体中残留和颤蚓体内累积的TBBPA含量均逐渐增加,其中高浓度组(1 mg·L-1)BCF值高于低浓度组(0.2 mg·L-1),两组平均BCF值为31.55,表明颤蚓对TBBPA的富集并不显著。
[Abstract]:In this paper, the toxic effects of tetrabromobisphenol A (TBBPA) on vermis and the enrichment effect of vermis on TBBPA were studied. First, 96 h-LC50 of TBBPA was measured by acute toxicity test, secondly, semi-static test was carried out according to the results of acute toxicity test. Six groups of 0 0. 025, 0. 25, 0. 25 and 1. 0 mg L ~ (-1) were selected for exposure test, and then 1 mg L ~ (-1) was cultured for 1 ~ (3) C ~ (5) for 7 and 10 days, and then 1 mg 路L ~ (-1) was cultured for 7 and 10 days. The enzyme activity in each group was determined, and the enrichment factor of TBBPA was determined after 7 days of exposure to TBBPA at high concentration (1 mg / L) and low concentration (0. 2 mg / L). The main results are as follows: 1. With the increase of exposure concentration and exposure time, the death rate of vermis increased and the toxicity of TBBPA was significantly increased. The LC50 of 48 h / 72 h / 96 h was 11.295 mg / L ~ (3.864) mg / L ~ (-1) and 0.22 mg / L ~ (-1), respectively, and the safe concentration was 0.22 mg / L ~ (-1). After exposure for 1 d and 7 d, the superoxide dismutase (SOD) in the vermis was induced, and the maximal induction was found in the 0.05 mg L-1 group, which was 1.36 and 1.61 times higher than that in the control group, and the activity of catalase CAT was mainly induced. The activity of CAT was lower than that of CAT at 7 days of exposure, and the activity of peroxidase (POD) showed the tendency of first induction and then inhibition, and the activity of P0. 01 and P0. 01% reached the peak value in the group of 0. 05 mg L-1, but the activity of CAT was lower than that of 1 day of exposure. In the 0.025 mg L-1 group, the maximum induction P0.01D was 1.28 and 1.38 times of that in the control group, and the activity decreased to the lowest in the 1 mg L-1 group, and the inhibition rates were 8% and 20%, respectively, and the acetylcholinesterase (ache) activity increased first and then decreased. The activity of AChE in each treatment group was significantly higher than that in the control group (P 0.01), and reached the peak value when the concentration of TBBPA was 0.25 mg L-1, which was 1.25 times and 1.20 times as much as that of the control group, respectively. The activities of cat and AChE increased first and then decreased within 10 days after exposure to 1 mg L-1TBBPA. The changes of cat and AChE activities reached the maximum on the 3rd and 5th day, respectively, and were decreased, increased, and then decreased. Its activity reached its peak value on the 3rd day (P0.01U. 4). With the increase of the concentration of venom, the TBBPA content of residual and vermis accumulated in water increased gradually, and the value of 1 mg L-1)BCF in the high concentration group was higher than that in the low concentration group, and the average BCF value of the two groups was 31.55, which indicated that the accumulation of TBBPA by the vermis was not significant.
