生长激素释放肽对大鼠脊髓缺血再灌注损伤的保护作用

【摘要】:目的探讨生长激素释放肽对大鼠脊髓缺血再灌注损伤的保护作用。方法将60只SD大鼠分为假手术组、损伤组、药物组,每组20只。损伤组和药物组以夹闭腹主动脉法制作大鼠缺血再灌注模型,假手术组不夹闭腹主动脉。术后,假手术组和损伤组大鼠腹腔注射生理盐水,药物组大鼠腹腔注射生长激素释放肽。分别于术后12、24、48和72 h取材,RT-PCR检测脊髓损伤区组织生长激素促分泌素受体-1α(GHSR-1α)mRNA的表达,酶联免疫吸附法测定血清肿瘤坏死因子-α(TNF-α)含量及髓过氧化物酶(MPO)活性,TUNEL法检测脊髓损伤区细胞凋亡情况,Western blot检测脊髓损伤区组织Bax、Bcl-2蛋白的表达。结果损伤组脊髓组织GHSR-1αmRNA表达量较假手术组明显下调(P0.05),药物组脊髓组织GHSR-1αmRNA表达量较损伤组明显上调(P0.05)。术后各时点,损伤组血清TNF-α含量及脊髓组织MPO活性均较假手术组明显增加(P0.05),而药物组血清TNF-α含量及脊髓组织MPO活性均较损伤组明显减小(P0.05)。术后24 h,药物组脊髓组织神经细胞凋亡率明显低于损伤组(P0.05)。术后各时点,药物组与损伤组脊髓组织Bax蛋白表达量差异均无统计学意义(P0.05),而术后24、48 h时药物组脊髓组织Bcl-2蛋白表达量均较损伤组上调(P0.01)。结论生长激素释放肽对大鼠脊髓缺血再灌注损伤具有保护作用,该作用与减少神经细胞炎症反应及凋亡等有关。
[Abstract]:Objective to investigate the protective effect of growth hormone releasing peptide (GH) on spinal cord ischemia reperfusion injury in rats. Methods Sixty SD rats were divided into sham-operation group, injury group and drug group with 20 rats in each group. The ischemia reperfusion model of rats was made by clamping abdominal aorta in injury group and drug group, and no clamping of abdominal aorta was observed in sham operation group. After operation, rats in sham operation group and injury group were intraperitoneally injected with normal saline, and rats in drug group were intraperitoneally injected with growth hormone releasing peptide. RT-PCR was used to detect the expression of growth hormone secretin receptor 1 伪 (GHSR-1 伪) mRNA in the injured area of spinal cord. The levels of serum tumor necrosis factor- 伪 (TNF- 伪) and the activity of myeloperoxidase (MPO) were determined by enzyme-linked immunosorbent assay (Elisa) and Tunel method was used to detect apoptosis in spinal cord injury area. Western blot was used to detect the expression of Baxfen Bcl-2 protein in spinal cord injury area. Results the expression of GHSR-1 伪 mRNA in spinal cord in the injured group was significantly lower than that in the sham operation group, and the expression of GHSR-1 伪 mRNA in the spinal cord in the drug group was significantly higher than that in the injured group. At all time points after operation, the content of TNF- 伪 in serum and the activity of MPO in spinal cord in the injured group were significantly higher than those in the sham operation group, while the content of TNF- 伪 in the serum and the MPO activity in the spinal cord in the drug group were significantly lower than those in the injured group. 24 hours after operation, the apoptotic rate of nerve cells in the spinal cord in the drug group was significantly lower than that in the injured group (P 0.05). At each time point after operation, there was no significant difference in the expression of Bax protein between the drug group and the injured group. However, at 24 h after operation, the expression of Bcl-2 protein in the drug group was significantly higher than that in the injured group (P 0.01). Conclusion growth hormone releasing peptide has protective effect on spinal cord ischemia-reperfusion injury in rats, which is related to the reduction of inflammatory reaction and apoptosis of nerve cells.
