纳米有机银簇化合物的设计合成及抑菌性研究

【摘要】:目的通过调控多金属氧酸盐阴离子的种类及尺寸,诱导合成系列纳米有机银簇抗菌剂;探索其抗菌活性,筛选出具有优良抗菌性能的抗菌剂。方法利用溶剂热方法合成系列纳米有机银簇化合物;通过X-射线单晶衍射、红外光谱、元素分析等方法对化合物进行结构表征。利用荧光光谱研究纳米有机银簇化合物的荧光特性,采用纳米无机材料抗菌性能检测方法研究试验化合物的抑菌活性。结果成功的合成出7个基于不同多金属氧酸盐模板剂的纳米有机银簇化合物。以发光型EuW_(10)多酸阴离子作为模板剂,首次合成出具有发光特性的42核(化合物1)和72核(化合物2)炔银簇化合物;以偏钒酸铵为模板剂原料,硫醇为有机配体,通过调节反应体系的pH,在甲醇溶剂中进行自组装反应,成功合成了系列硫醇银纳米簇合物(3~7)。其中,化合物4~6是首次在纳米尺度上合成出具有柏拉图模型的正十二面体有机银簇化合物,也是首例具有酸碱诱导变色的纳米银簇化合物。该类化合物的成功合成不仅为纳米银簇家族添加了新成员,也为对外界刺激有应激反应的智能型簇合物的研究提供新的结构模型。对大肠杆菌抑菌活性试验表明,化合物1和2抑菌活性良好,抑菌率分别为95.5%和97.7%。化合物3~7的抑菌性显著不同,抑菌率分别为21.4%,70.0%,95.1%,81.6%和59.2%。结论以叔丁基炔或叔丁基硫醇为有机配体,通过溶剂热法设计合成了7个新型纳米有机银簇化合物,研究纳米银簇对大肠杆菌的抑菌性能。抑菌实验结果表明,纳米有机银簇化合物的抗菌效果明显高于前躯体叔丁基炔银和硫醇银;另外,纳米有机银簇化合物核心模板剂(多酸阴离子)种类和构型,以及银壳层中银核数量和构型的不同都会显著影响化合物对大肠杆菌的抑制作用。该类化合物的成功制备,为有机银簇在抗菌领域的应用提供了基础数据和结构模型。
[Abstract]:Objective to induce the synthesis of a series of nano-organosilver cluster antimicrobial agents by regulating the species and size of polyoxometalate anions, to explore their antibacterial activities and to screen out antimicrobial agents with excellent antibacterial properties. Methods A series of nano-organosilver cluster compounds were synthesized by solvothermal method and characterized by X-ray single crystal diffraction, infrared spectroscopy and elemental analysis. Fluorescence spectra were used to study the fluorescence characteristics of nano-organo silver cluster compounds, and the antibacterial activity of the tested compounds was studied by the method of antimicrobial properties of nano-inorganic materials. Results Seven nanoorganosilver clusters based on different polyoxometalate templates were successfully synthesized. Silver clusters of 42 nuclei (compounds 1) and 72 nuclei (compounds 2) with luminescent properties were synthesized by using the luminescent EuWS 10) polyacid anions as templates, using ammonium metavanadate as template material and mercaptan as organic ligands. A series of silver mercaptan nanoclusters were successfully synthesized by self-assembly reaction in methanol solvent by adjusting the pH of the reaction system. Among them, compound 4N 6 is the first organic dodecahedron silver cluster compound with Plato model on nanometer scale, and it is also the first nano-silver cluster compound with acid-base induced color change. The successful synthesis of these compounds not only adds new members to the silver nanoclusters, but also provides a new structural model for the study of intelligent clusters with external stimuli. The bacteriostatic activity of compounds 1 and 2 was good in Escherichia coli. The bacteriostatic rates of compounds 1 and 2 were 95. 5% and 97. 7%, respectively. The antimicrobial activity of compound 3Y7 was significantly different, the antibacterial rate was 21.40.0and 95.1% and 59.2%, respectively. Conclusion Seven novel nano-organosilver clusters were synthesized by solvothermal method using tert Ding Ji acetylene or tertiary Ding Ji mercaptan as organic ligands, and the bacteriostatic properties of nano-silver clusters against Escherichia coli were studied. The results of bacteriostatic test showed that the antibacterial effect of nano-organo silver cluster compounds was significantly higher than that of tert-tert Ding Ji silver and mercaptan silver, in addition, the types and configurations of core templates (polyacid anions) of nano-organo silver clusters were higher than those of precursor tert-silver clusters. The number and configuration of silver nuclei in the silver shell significantly affected the inhibition of the compounds on Escherichia coli. The successful preparation of this kind of compounds provides the basic data and structural model for the application of organic silver clusters in antibacterial field.
