美索巴莫磷酸钠对小鼠抗惊厥和肌松作用的研究

【摘要】:目的研究美索巴莫磷酸钠对小鼠的抗惊厥作用及肌肉松弛作用。方法将小鼠按照体重随机分组,每组10只,雌雄各半,将美索巴莫磷酸钠(Me-Na)制备成质量分数为231,462,924,1848 mg·kg~(-1)的系列溶液,采用灌胃(给药后30 min实验)和注射(给药后10 min实验)两种途径给予小鼠,并与美索巴莫(650 mg·kg~(-1))作用效果对比。抗惊厥实验:分别研究不同浓度美索巴莫磷酸钠抗电惊厥及抗士的宁惊厥作用,给药后记录小鼠的惊厥发生率和惊厥出现时间,研究其抗惊厥效果。肌肉松弛实验:给药后通过检测小鼠的抓力和转杆时间,研究美索巴莫磷酸钠对小鼠的肌肉松弛作用。结果抗电惊厥实验:灌胃组中的空白组、对照组、实验Ⅱ、Ⅲ、Ⅳ组发生例数为10,0,6,4,0例;注射组中的空白组、对照组、实验Ⅲ组发生例数为10,3,5例,与空白组相比,对照组和实验组的差异有统计学意义(P0.05)。抗士的宁惊厥实验:灌胃组中的空白组和实验Ⅱ组惊厥出现的时间为(8.89±1.69),(14.60±3.81)min,注射组中的空白组、对照组和实验Ⅰ、Ⅱ、Ⅲ组惊厥出现的时间为(6.44±1.86),(12.31±2.34),(19.88±3.79),(12.73±1.72),(13.22±1.67)min,与空白组相比,对照组和实验组的差异有统计学意义(P0.01)。抓力实验:灌胃组中的空白组、对照组和实验Ⅱ、Ⅲ、Ⅳ组的抓力与体重的比值分别为(6.44±0.54),(3.14±0.97),(4.18±1.31),(3.51±1.65),(2.93±1.06)N·g~(-1),注射组中的空白组、对照组和实验Ⅰ、Ⅱ、Ⅲ组的抓力与体重的比值为(6.67±0.69),(1.54±0.33),(2.84±0.53),(3.83±0.92),(3.22±1.09)N·g~(-1),与空白组相比,对照组和实验组的差异有统计学意义(P0.01)。转杆实验:灌胃组中的空白组、对照组和实验Ⅲ、Ⅳ组的转杆持续时间为(175.24±7.08),(5.96±4.84),(10.83±6.27),(3.73±1.66)s,注射组中的空白组、对照组和实验Ⅱ、Ⅲ组转杆持续时间为(174.71±7.29),(2.33±0.58),(7.06±4.45),(1.56±0.62)s,与空白组相比,对照组和实验组的差异有统计学意义(P0.001)。结论美索巴莫磷酸钠对小鼠具有抗惊厥作用和肌肉松弛作用,且与临床应用的美索巴莫效果相当。
[Abstract]:Objective to study the anticonvulsant and muscle relaxation effects of sodium mesophosphate on mice. Methods mice were randomly divided according to their body weight, 10 mice in each group, half male and half female. A series of solutions with mass fraction of 231462U 924m 1848 mg 路kg ~ (-1) were prepared from Me-Na. Mice were given intragastric administration (30 min after administration) and injection (10 min after administration), and the effects were compared with that of misobutamol (650 mg / kg).
