HepG2.2.15细胞的差异性表达及对IFN-α的反应性研究

【摘要】:采用有限稀释法从HepG2.2.15细胞株中分离出10株单克隆细胞株。ELISA法检测其HBsAg与HBe Ag表达量,根据HBsAg、HBe Ag表达量的不同,筛选出最高、中等、最低3株克隆细胞株。MTT法进行干扰素(IFN)-α的细胞毒作用研究,表明IFN-α对不同克隆细胞株药物毒作用相似。ELISA法检测各克隆细胞株给IFN-α前后HBsAg、HBe Ag表达量,IFN-α对HepG2.2.15细胞HBe Ag的抑制作用较弱,对HBsAg、HBV DNA的抑制作用较强,且对表达量高的克隆细胞株抑制作用更强。不同HepG2.2.15细胞表达HBsAg与HBe Ag存在差异性,且表达量高的细胞株对治疗浓度的IFN-α更加敏感。
[Abstract]:The expression of HBsAg and HBe Ag in HepG2.2.15 cell line was detected by Elisa method. The HBsAg and HBe Ag expression levels were determined by using the limited dilution method. According to the different expression levels of HBe Ag in HBsAg, the highest and moderate HBe Ag expression levels were obtained from HepG2.2.15 cell line. The cytotoxicity of IFN- 伪 was studied by MTT assay. The results showed that IFN- 伪 had similar drug toxicity to different clone cell lines. Elisa method was used to detect the expression of HBsAg HBe Ag in HBe cells before and after IFN- 伪 was given. IFN- 伪 had a weak inhibitory effect on HBe Ag in HepG2.2.15 cells, and a stronger inhibitory effect on HBe DNA in HBN- 伪 cells before and after IFN- 伪 administration. Moreover, it had stronger inhibitory effect on clone cell lines with high expression. Different HepG2.2.15 cells expressed HBsAg and HBe Ag differently, and the cell lines with high expression were more sensitive to IFN- 伪.
