人血高铁血红蛋白浓度快速检测卡的研制

【摘要】:目的: 明确不同浓度亚硝酸钠溶液与高铁血红蛋白生成率间的量效关系方程,采用RGB色彩模式,研制出一种基于比色卡原理技术的、快速、准确、安全且经济实用的人血高铁血红蛋白浓度检测卡。 方法: 利用不同浓度亚硝酸钠溶液孵育健康成人献血者血样,得到亚硝酸钠溶液与高铁血红蛋白生成率间的量效关系方程。根据亚硝酸钠溶液与高铁血红蛋白生成率间的量效关系方程,获取浓度百分比为10%~80%(间隔10%)高铁血红蛋白血样所需的不同浓度亚硝酸钠溶液。 将健康献血者未经孵育血样与浓度百分比为10%~80%(间隔10%)高铁血红蛋白血样分别滴加在血样采集卡上,并用RGB ColorFinder软件扫描获取红绿蓝(red, green, blue, RGB)值和颜色十六进制代码(HEX Code),最终生成相应的高铁血红蛋白色块,制成检测卡。 结果: 亚硝酸钠溶液(X)与高铁血红蛋白生成率(Y)间的量效关系方程为:Y=2.12598+0.7123X(R2=0.9982,P0.01),,亚硝酸钠溶液与高铁血红蛋白生成率间呈线性相关。当需形成浓度百分比为10%~80%(间隔10%)高铁血红蛋白时,亚硝酸钠溶液浓度分别为11mmol/L、25mmol/L、39mmol/L、53mmol/L、67mmol/L、81mmol/L、95mmol/L、109mmol/L。 根据健康成人献血者血样及浓度百分比为10%~80%(间隔10%)高铁血红蛋白血样获取的RGB值分别为:(205.10.16)、(193.33.35)、(183.34.38)、(170.35.39)、(157.36.41)、(144.37.43)、(133.38.44)、(119.39.48)、(103.40.49);HEX Code值为(#CD0A10)、(#C12123)、(#B7226)、(#AA2327)、(#9D2429)、(#90252B)、(#85262C)、(#772730)、(#672831)。与健康成人献血者血样相比,随着高铁血红蛋白浓度百分比的增高,血样采集卡上的颜色逐渐变深至棕褐色。 结论: 本实验所制作而成的人血高铁血红蛋白浓度快速检测卡,为法医日常检案、刑事侦查及临床医务工作者检测人血高铁血红蛋白的浓度提供了快速、准确、安全且经济实用的检测方法,值得推广使用。
[Abstract]:Objective: to determine the dose-effect relation equation between sodium nitrite solution and methemoglobin production rate in different concentrations, and to develop a rapid and accurate method based on the principle of colorimetric card by using RGB color model. Safe and economical human blood methemoglobin concentration test card. Methods: blood samples of healthy adult blood donors were incubated with different concentrations of sodium nitrite solution, and the dose-effect relationship equation between sodium nitrite solution and methemoglobin production rate was obtained. According to the dose-effect relation equation between sodium nitrite solution and methemoglobin production rate, the different concentrations of sodium nitrite solution needed for the blood sample of methemoglobin were obtained. Add the unincubated blood samples and the blood samples of the healthy donors to the blood sample collection card with a percentage of 10% and 80% (interval 10) of methemoglobin, The RGB ColorFinder software was used to scan red, green, blue, RGBand hexadecimal codes of HEX code. Finally, the corresponding methemoglobin color blocks were generated, and the detection cards were made. Results: the equation of dose-effect relationship between sodium nitrite solution (X) and methemoglobin production rate (Y) was: 1: Y2.12598 0.7123XC R2O0.9982P0.01C, and there was a linear correlation between sodium nitrite solution and methemoglobin production rate.
