真皮内注射二甲氨基乙醇复合制剂对衰老大鼠皮肤胶原代谢及相关通路分子表达的影响

【摘要】:目的:研究真皮内注射二甲氨基乙醇(DMAE)复合制剂对亚急性衰老大鼠皮肤胶原代谢及TGFβ1/smads通道的影响。方法:30只Wistar大鼠随机分入衰老生理盐水组(A组)、衰老治疗组(B组)、正常对照组(C组),各10只。A、B两组颈背部皮下注射D-半乳糖125mg/kg/d 42天。从第18天开始于A、B组臀背部分别注射生理盐水和DMAE复合制剂1ml,每周一次,连续四周。每次治疗后3天进行活体皮肤共聚焦纤维镜检测。42天处死所有大鼠,取注射药物区域皮肤,观察各组皮肤真皮厚度、胶原纤维密度和皮肤羟脯氨酸(HYP)含量的变化,RT-PCR法检测皮肤组织TGFβ1、smad3、Ⅰ、Ⅲ型前胶原(colⅠcolⅢ)脂质金属蛋白酶-1(MMP-1)、脂质金属蛋白酶抑制剂-1(TIMP-1)m RNA表达量,以及免疫组化检测成纤维细胞增殖细胞核抗原(PCNA)和TGFβ1的表达。结果:衰老治疗组与衰老生理盐水组大鼠相比真皮厚度、胶原纤维密度、羟脯氨酸含量及TIMP-1 m RNA表达量显著增加,皮肤组织colⅠ、TGFβ1、smad3 m RNA表达量增加(P0.05),而对colⅢ、MMP-1 m RNA及PCNA表达量没有影响。结论:真皮内注射DMAE复合制剂(赛雷弗#174;)可正调节衰老皮肤中成纤维细胞TGFβ1/smad3基因表达,促进Ⅰ型胶原蛋白合成,增加衰老皮肤中胶原的含量和真皮厚度,且通过调节MMP-1/TIMP-1平衡抑制胶原蛋白降解发挥其抗衰老作用。

From the 18th day, 1 ml of saline and DMAE were injected into the buttock back of group A and B respectively, once a week for four weeks. Three days after each treatment, all rats were killed by confocal fibroscope examination of living skin on day 42, and the dermal thickness of each group was observed by injecting drug area skin.
