大鼠中抗凝血化合物TI-02的含量测定研究

【摘要】:对一类肽化合物的抗凝活性进行初步筛选,同时建立测定大鼠血浆中化合物TI-02浓度的HPLC方法。选用C18柱(250 mm×4.6 mm,10μm),采用甲醇-乙腈-水三元体系梯度洗脱,流速1.0 mL·min~(-1),检测波长240 nm,柱温30℃,进样量20μL。抗凝血化合物TI-02在浓度0.125~10.0μg/mL范围内线性良好,r=0.9964;回收率为97.1%~106.7%;日内、日间精密度RSD2.3%,室温24 h内稳定,RSD1.0%。本实验建立HPLC测定方法灵敏度高、精密度好,结果稳定可靠,可用于该肽类化合物大鼠血浆中浓度的测定。
