用HPLC法测定人血浆中丙戊酸钠的游离浓度

【摘要】:目的建立一种测定人血浆中丙戊酸钠游离浓度的中空纤维离心超滤-高效液相色谱(HPLC)方法。方法血浆样本经中空纤维离心超滤技术预处理,经2-溴甲基-2-萘乙酮衍生后进行HPLC分析。色谱柱:Diamonsil RPC18(4.6 mm×150 mm,5μm),流动相:甲醇∶水=80∶20;流速:1.0 m L·min~(-1);柱温:30℃;检测波长:251 nm。考察该方法的专属性、标准曲线和定量下限、精密度与回收率、稳定性。结果人血浆中游离的丙戊酸钠在该条件下分离良好,在0.05~50.00μg·m L~(-1)内线性关系良好(r=0.999 0),定量下限为0.01μg·m L~(-1),方法回收率为96.51%~101.22%,绝对回收率达到94.28%~99.15%,批内、批间RSD均4.00%,在室温放置10 h、冻融循环3次和冰冻21 d的稳定性良好。结论本方法简便、准确、灵敏,适用于人血浆中丙戊酸钠的游离浓度分析。
