雷帕霉素互变异构体的分离和鉴定

【摘要】:目的分离、纯化HPLC图谱中一直伴随着雷帕霉素的异构体(Rap-T)和它的转化产物并鉴定它们的化学结构。方法制备液相色谱仪分离Rap-T及其转化产物;HPLC跟踪分析Rap-T在25℃、50%乙腈-水溶液中不同时间的转化结果 ;对Rap-T及其转化产物进行理化性质和波谱分析。结果 Rap-T是雷帕霉素化学结构上的吡喃环扩展为氧杂环庚烷的化合物,其转化产物的化学结构与雷帕霉素一致;Rap-T与雷帕霉素在25℃、50%乙腈-水溶液中能相互转化。结论 Rap-T是雷帕霉素的互变异构体,二者可以相互转化。
