载纳米磺胺嘧啶银明胶凝胶体外抗菌及体内促愈合性能

【摘要】:目的制作包载纳米磺胺嘧啶银(AgSD)的京尼平交联明胶凝胶,考察其体外抗菌及体内促愈合性能。方法分别配制纳米AgSD与粗粉AgSD梯度浓度混悬液、含纳米AgSD与粗粉AgSD的明胶交联凝胶和不含AgSD的空白凝胶。测量纳米AgSD凝胶对敏感细菌的抑菌圈直径;MTT法考察其对L929成纤维细胞的相容性,测定细胞相对生长率;考察纳米AgSD混悬液对3种敏感细菌的最小抑菌浓度(MIC)和最小杀菌浓度(MBC);建立小鼠深Ⅱ度烫伤合并金黄色葡萄球菌感染模型,应用载药凝胶敷料治疗3周后计算愈合指数,天狼星红染色考察小鼠创面胶原沉积,HE染色观察创面组织病理学变化并评分。结果在抑菌圈、MIC和MBC测定实验中,纳米AgSD较粗粉混悬液抗菌性能明显提高,且载纳米AgSD凝胶较市售乳膏有更好的抗菌性及细胞相容性;小鼠创面体内愈合实验中,从给药第2周开始纳米AgSD凝胶组创面愈合指数较涂菌组、乳膏组和粗粉凝胶组显著提升(P0.05);且与对照组相比,小鼠创面Ⅰ型胶原、总胶原含量及组织病理学评分均明显升高(P0.05)。结论载纳米AgSD明胶凝胶具有良好的体外抗菌效果及快速促机体愈合性能。
[Abstract]:Aim to prepare a genipine crosslinked gelatin gel coated with silver sulfadiazine (AgSD) and investigate its antibacterial and healing properties in vitro. Methods the gradient concentration suspensions of nanometer AgSD and crude AgSD, gelatin crosslinked gel containing nanometer AgSD and blank gel without AgSD were prepared respectively. The diameter of bacteriostasis circle of nanometer AgSD gel on sensitive bacteria was measured, the compatibility of L929 fibroblasts and the relative growth rate of L929 fibroblasts were investigated by MTT method. The minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC);) of nanometer AgSD suspension against three sensitive bacteria were investigated to establish the model of deep second degree scald with Staphylococcus aureus infection in mice. The healing index was calculated after 3 weeks of treatment with drug loaded gel dressing. Sirius red staining was used to study the collagen deposition in the wound and HE staining was used to observe the histopathological changes of the wound. Results in the test of bacteriostatic circle, MIC and MBC, the antibacterial activity of nanometer AgSD was significantly higher than that of crude powder suspension, and the antibacterial and cytocompatibility of nano-AgSD gel was better than that of marketable cream.
