血液和尿液中吲哚美辛和布洛芬含量测定的方法学研究及其应用

【摘要】:目的:非甾体抗炎药(Nonsteroidal Antiinflammatory Drugs,NSAIDs)因为有解热、降低疼痛、对抗炎症和对抗风湿等临床作用而被广泛的应用于临床。本实验拟建立一种同时测定血浆和尿液中吲哚美辛和布洛芬的药物浓度的HPLC方法。并运用该方法对实际样品进行测定,考察两种药物的血药浓度变化情况。方法:ZORBAX SB-C18柱(4.6mm×250mm,5μm)色谱柱,流动相为乙腈和醋酸铵溶液(20mmol/L,pH=3.5)=85:15(v/v),流速为1mL/min,进样量80μL,布洛芬保留时间为8.57min左右,吲哚美辛的保留时间6.78min左右,柱温为室温,检测波长220nm。结果:吲哚美辛和布洛芬能够与内源性物质有效分离,在较宽的浓度范围内吲哚美辛和布洛芬的峰面积和与其浓度呈较好的线性关系(R"g0.9985),方法的精密度小于4.0%,方法回收率大于85%。生物样品在4℃冰箱中放置48小时性质稳定。结论:本方法专属性好,实验所用时间短。可作为生物样品中吲哚美辛和布洛芬的药物浓度测定的方法。运用此方法对数只大鼠血药浓度监测结果表明,布洛芬在1h左右达到最大血药浓度(27.65μg/mL),吲哚美辛在3h左右达最大血药浓度(2.71μg/mL)。本研究为进一步研究吲哚美辛和布洛芬体内利用度奠定基础,为临床安全合理用药提供了依据。也为其他非甾体抗炎药的分析提供了参考。
[Abstract]:Objective: Non-steroidal anti-inflammatory drug (Nonsteroidal Antiinflammatory Drugs,NSAIDs) has been widely used in clinic because of its antipyretic, pain reduction, anti-inflammatory and anti-rheumatic effects. A HPLC method for simultaneous determination of indomethacin and ibuprofen in plasma and urine was developed. The actual samples were determined by this method, and the changes of blood concentration of the two drugs were investigated. Methods: ZORBAX SB-C18 column (4.6mm 脳 250mm ~ 5 渭 m) was used. The mobile phase consisted of acetonitrile and ammonium acetate solution (20 mmol / L ~ (-1) pH ~ (-1) = 85:15 (v / v), the flow rate was 1 mL / min, the injection amount was 80 渭 L, and the retention time of ibuprofen was about 8.57min. The retention time of indomethacin was about 6.78min, the column temperature was room temperature and the detection wavelength was 220 nm. Results: indomethacin and ibuprofen could be effectively separated from endogenous substances. The peak area and concentration of indomethacin and ibuprofen were linear in a wide range of concentrations (R "g 0.9985).
