舍曲林诱导Kasumi-1细胞凋亡及其作用机制的实验研究

【摘要】:目的:探讨精神类药物舍曲林对人急性髓系白血病细胞株Kasumi-1的生物学效应。方法:将不同浓度舍曲林作用于Kasumi-1细胞不同时间,通过细胞计数、细胞形态学检查、流式细胞术和Western blot分别检测舍曲林处理前后Kasumi-1细胞增殖和凋亡情况、细胞周期分布以及相关蛋白的改变。结果:舍曲林能够抑制Kasumi-1细胞增殖、诱导细胞凋亡;15和20μmol/L舍曲林处理Kasumi-1细胞24 h后,生长抑制率分别达到(19.00±7.37)%和(47.90±11.19)%;流式细胞术检测结果显示,与空白对照组相比,15和20μmol/L舍曲林处理Kasumi-1细胞24 h后,Annexin V阳性细胞百分比从(9.71±2.12)%分别上升至(20.54±2.52)%和(45.37±7.88)%(P0.01),且细胞被明显阻滞在G0/G1期和G2/M期;此外,舍曲林还可以下调细胞内翻译控制肿瘤蛋白(translationally controlled tumor protein,TCTP)。结论:舍曲林能够显著诱导Kasumi-1细胞发生凋亡,其作用机制可能与细胞内TCTP蛋白的下调相关。
[Abstract]:Objective: to investigate the biological effects of sertraline on human acute myeloid leukemia cell line Kasumi-1. Methods: Kasumi-1 cells were treated with Sertraline at different concentrations for different time. Cell count, cell morphology, flow cytometry and Western blot were used to detect the proliferation and apoptosis of Kasumi-1 cells before and after sertraline treatment. Cell cycle distribution and changes in related proteins. Results: Sertraline inhibited the proliferation of Kasumi-1 cells and induced apoptosis, and the growth inhibition rates of Kasumi-1 cells treated with sertraline for 24 h were (19.00 卤7.37)% and (47.90 卤11.19)%, respectively. The results of flow cytometry showed that Kasumi-1 cells were treated with 15 and 20 渭 mol/L sertraline for 24 h, compared with the control group. The percentage of Annexin V positive cells increased from (9.71 卤2.12)% to (20.54 卤2.52)% and (45.37 卤7.88)%, respectively. In addition, sertraline can also down-regulate intracellular translation control tumor protein (translationally controlled tumor protein,TCTP). Conclusion: Sertraline can induce apoptosis in Kasumi-1 cells, and its mechanism may be related to the down-regulation of TCTP protein.
