多功能复合载药脂质体的制备方法及性能

【摘要】:以天然大豆卵磷脂为主要材料,胆固醇、近红外量子点CdSeTe和超顺磁性Fe_3O_4粒子为附加材料,DO_X为包封的药物,制备了多功能复合载药脂质体。通过正交试验选取影响复合载药脂质体粒径和包封率的5个因素,即磷脂浓度、药脂比、超声时间、水化温度和Fe_3O_4量,每个因素选取4个水平进行优化实验。根据最优条件,采用薄膜分散发制备了多功能复合载药脂质体并对其粒径、磁性和荧光性质进行了测定。结果表明多功能复合载药脂质体粒径大约200nm、近似球形,且具有良好的近红外荧光性能和磁响应性能。
[Abstract]:Using natural soybean lecithin as main material, cholesterol, near-infrared quantum dot CdSeTe and superparamagnetic Fe_3O_4 particles as additional materials, and DO_X as encapsulated drug, the multifunctional compound drug-loaded liposomes were prepared. Five factors affecting the particle size and entrapment efficiency of the composite liposome were selected by orthogonal test, namely, the concentration of phospholipid, the ratio of liposomes to liposomes, ultrasonic time, hydration temperature and the amount of Fe_3O_4, and each factor was selected at 4 levels to optimize the experiment. According to the optimum conditions, the multifunctional composite liposomes were prepared by dispersion of films and their particle size, magnetic properties and fluorescence properties were determined. The results show that the multifunctional composite liposomes are about 200 nm in diameter, nearly spherical, and have good near infrared fluorescence and magnetic response.
