高效液相色谱法检测丙泊酚及依托咪酯混合液化学稳定性的研究

【摘要】:目的采用高效液相色谱法(HPLC)体外模拟体内环境条件确定丙泊酚、依托咪酯混合后的新物质生成情况。方法采用十八烷基硅烷键合硅胶作为填充剂(250.0mm×4.6mm,5μm),以磷酸盐缓冲液-乙腈(调至pH7.4)作为流动相[磷酸盐缓冲液∶乙腈=35∶65(V/V)],设定柱温为30℃,流速为1.0mL/min,进样量为10μL,全波长下进行紫外光谱检测(波长190~800nm),通过对比标准品及对照品图谱确认图谱中是否有新的特征峰。结果通过与标准品图谱进行比较发现,丙泊酚、依托咪酯二者混合品在体外模拟体内环境条件下无新的特征峰出现,即未生成第3种物质。结论所建方法简便、快速、准确地检测出丙泊酚、依托咪酯混合后无新物质生成。
