新疆马脂不饱和脂肪酸软胶囊的制备工艺研究

【摘要】:目的:采用压榨法及溶剂结晶法对马脂不饱和脂肪酸进行富集提取,其主要功效成分为油酸及亚油酸,并筛选添加抗氧化剂,研制马脂不饱和脂肪酸软胶囊。方法:1.以酸值、碘值、过氧化值及产率为考察指标,采用单因素试验,优选马油毛油的提取工艺。2.采用溶剂结晶法对马脂不饱和脂肪酸进行提取,对所得油脂气相色谱质谱联用技术分析马脂脂肪酸成分,以油酸综合产量为考察指标,通过单因素试验筛选最佳溶剂种类及最佳提取次数并采用正交试验考察结晶温度、结晶时间及混合脂肪酸与溶剂比例对马脂不饱和脂肪酸溶剂结晶的影响。3.以马脂不饱和脂肪酸作为软胶囊主成分,采用单因素考察法筛选抗氧化剂种类及添加量,确定软胶囊制备工艺。结果:1.压榨法影响因素中不同提取时间的影响比不同提取温度的影响更大;最佳提取工艺为:提取时间6h,提取温度80-C;此条件下数据结果最为理想,产率为39.44±0.07g,酸值碘值过氧化值分别为3.580mg/g,81.012g/100g,0.916mmol/kg。2.溶剂结晶提取工艺:通过单因素试验得到:最佳溶剂是95%乙醇,最佳工艺条件是结晶温度2。C,最佳结晶时间6h,最佳混合脂肪酸与溶剂比例是1:7;滤饼进行重复结晶的最佳次数为两次;通过正交试验得到最佳的工艺条件是:混合脂肪酸与溶剂比例是1:9;结晶时间为8h;结晶温度为2。C。3.添加抗氧化剂的最佳种类为维生素E,最佳添加量为0.02%,软胶囊囊壳中的成分明胶甘油水的最佳比例为1:0.5:1。结论:压榨法相对溶剂提取法更方便、安全,且产率较高,适合大规模工业生产;溶剂结晶法提取后不饱和脂肪酸含量较提取前提高了24.39%,油酸含量提高了20.21%。经溶剂结晶法提取后的马油,含有更高的单不饱和脂肪酸,可开发为极具潜力的保健品。
[Abstract]:Objective: to enrich and extract the unsaturated fatty acids of equine by squeezing and solvent crystallization. The main components are oleic acid and linoleic acid. The soft capsules of unsaturated fatty acids of equine fat were prepared by screening and adding antioxidants. Methods: 1. With acid value, iodine value, peroxide value and yield as the index of investigation, single factor test was used to optimize the extraction process of horse oil wool oil. 2. The unsaturated fatty acids of horse fat were extracted by solvent crystallization method. The fatty acids were analyzed by GC-MS. The comprehensive yield of oleic acid was used as the index. The effects of crystallization temperature, crystallization time and the ratio of mixed fatty acids to solvent on the solvent crystallization of unsaturated fatty acids were investigated by orthogonal test. Taking the unsaturated fatty acid of horse fat as the main component of soft capsule, the kinds of antioxidants and the amount of antioxidants were selected by single factor investigation method, and the preparation process of soft capsule was determined. Results: 1. The influence of different extraction time was greater than that of different extraction temperature, and the optimum extraction technology was as follows: extraction time 6 h, extraction temperature 80-C; Under this condition, the yield is 39.44 卤0.07g, the peroxide value of acid value iodine is 3.580mg / g / 100g / 100mg / 100mg 路kg ~ (-2), and the value of peroxide value is 3.580mg / g / 100g / 100g / 100g / 100g ~ (-1) 路g ~ (-1), respectively. Conclusion: the squeezing method is more convenient, safe and suitable for large-scale industrial production than solvent extraction, and the content of unsaturated fatty acids and oleic acid after solvent crystallization is 24.39 and 20.21 respectively. The horse oil extracted by solvent crystallization method contains higher monounsaturated fatty acids and can be developed as a potential health product.
