1-溴丙烷对大鼠大脑线粒体损伤及依达拉奉保护作用的观察

【摘要】:目的观察1-溴丙烷(1-BP)对大脑线粒体损伤和依达拉奉(EDA)的保护作用。方法将成年雄性Wistar大鼠随机分为对照组、1-BP(800 mg/kg·bw)组、1-BP+1 mg/kg·bw EDA组、1-BP+3 mg/kg·bw EDA组、1-BP+5 mg/kg·bw EDA组、5 mg/kg·bw EDA对照组,每组12只。分别经灌胃和腹腔注射给予1-BP和EDA,1次/d,连续12 d。动物处死后,迅速剥离大脑,采用差速离心法提取大脑前额叶皮层线粒体,检测细胞线粒体复合体Ⅰ、Ⅱ、Ⅲ和Ⅳ活性;测定还原型谷胱甘肽(GSH)和氧化型谷胱甘肽(GSSG)含量,并计算GSH/GSSG比值;采用抗Neu N抗体进行免疫组化染色,计数神经元。结果 1-BP明显降低大脑前额叶皮层细胞线粒体复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ活性(P0.05,P0.01);1-BP组动物大脑皮层GSH含量也较对照组降低(P0.01),GSSG含量升高(P0.01),GSH/GSSG比值明显降低(P0.01);给予1-BP染毒后,大脑前额叶皮层神经元明显减少(P0.01)。1-BP染毒同时给予不同剂量的EDA,大鼠大脑前额叶皮层线粒体复合体活性均有不同程度的升高;GSH含量降低也被明显逆转,GSH/GSSG比值相应升高(P0.05,P0.01);神经元丢失明显改善(P0.05,P0.01);EDA对1-BP诱导大脑损伤的保护作用呈现剂量-效应关系(P0.05)。结论 1-BP导致大脑皮层线粒体损伤,脑内氧化应激及神经元丢失,EDA可有效减轻1-BP神经毒性。
[Abstract]:Objective to observe the protective effect of 1-bromopropane (1-BP) on cerebral mitochondria injury and Edaravone (EDA). Methods Adult male Wistar rats were randomly divided into control group, 1-BP (800 mg/kg bw) group, 1-BP 1 mg/kg bw EDA group, 1-BP 3 mg/kg bw EDA group and 1-BP 5 mg/kg bw EDA group. 5 mg/kg bw EDA control group, 12 rats in each group. 1-BP and EDA,1 were given intraperitoneally and intraperitoneally respectively for 12 days.
